VEGF-A对人肝癌细胞体外形成血管生成拟态能力的影响

         

摘要

目的 探讨血管内皮生长因子(VEGF)对人肝癌细胞体外形成血管生成拟态(VM)能力的影响.方法 采用不同浓度的VEGF-A诱导人肝癌PLC细胞,分别用划痕实验、明胶酶谱法分析不同浓度VEGF-A对PLC细胞迁移性和侵袭性的影响;三维培养观察VEGF-A对PLC细胞形成VM能力的影响;Western blot法检测VEGF-A处理后的血管内皮钙黏附素( VE-cadherin)表达.结果 VEGF-A呈剂量依赖性增强肝癌PLC细胞的迁移率和侵袭相关蛋白MMP-9的活性,促进管道样结构形成及VE-cadherin表达(P均<0.05).VEGF-A水平与管道数量呈正相关(r =0.929,P<0.05).结论 VEGF-A可增强肝癌PLC细胞的迁移和侵袭能力,促进其VM形成,使肿瘤组织血供丰富,促进肿瘤浸润和转移.%Objective To study the effects of vascular endothelial growth factor (VEGF) on vasculogenic mimicry (VM) of human hepatocellular carcinoma cells in vitro. Methods Hepatocellular carcinoma cells PLC were induced with various concentrations of VECF-A. The migration and invasion abilities of PLC when treated with various concentrations of VEGF-A were detected by wound-healing and Gelatin zymography assay. The effects of VEGF-A on vasculogenic mimicry in PLC cells were detected in three-dimensional cultures, and the expressions of vascular endothelial cadherin ( VE-cadherin) were detected after treated with VEGF-A by Western blot. Results VEGF-A could enhance the ability of the migration, and the activities of invasion-related protein MMP-9 in dose-dependent manner, and also promote the VM formation and the expression of VE-cadherin of PLC cells (P < 0. 05). The correlation between the level of VEGF-A and the quantities of the pipelines were positive significantly (r =0.929, P<0.05). Conclusion VEGF-A can promote the migration, invasion and the ability of VM formation in the hepatocellular carcinoma cells PLC, and then promote the blood supply of tumor, and the invasion and metastasis of tumor.
