Objective To observe the expression of osteopontin( OPN) in the largngeal squamous cell carcinoma (LSCC) tissues, and the effect on the proliferation and invaaiveness of laryngeal cancer Hep-2 cells of lentivirus vector with interference OPN ( LV-shOPN). Methods SP immunohistochemical method was used to detect the OPN of 44 LSCC and adjacent tissues. LV-shOPN was transfecled into Hep-2 cells. OPN in the Hep-2 cells was detected by Western blot was detected by Transwell assay, the proliferation of Hep-2 cells was examined by MTT. The invasion ability of Hep-2 cells was detected by Transwell assay. Results The positive rate of OPN in LSCC and adjacent tissue was 75. 0 % (33/44) and 13. 6% {6/44)respectively, the two rates were compared, P <0.01. The OPN expression levels were significantly correla ted with the presence of lymph node, clinical stage and histopathology grade of LSCC, all P <0.01. After LV-shOPN trans fection, the expression of OPN was reduced and ability of the proliferation and invasiveness was decreased in Hep-2 cells (all P<0.05). Conclusions OPN expression is higher in LSCC than that in adjacent tissue, and maybe participated in the development and metastasis of LSCC. LV-shOPN can inhibit efficiently the proliferation and invasiveness of Hep-2 cells.%目的 观察人喉鳞状细胞癌(LSCC)组织中骨桥蛋白(OPN)的表达变化及慢病毒shOPN对喉癌Hep-2细胞侵袭转移的影响.方法 采用免疫组化SP法,检测44例LSCC及癌旁正常组织中的OPN.干扰OPN慢病毒载体(LV-shOPN)转染Hep-2细胞,Western blot检测该细胞其中的OPN,MTT法检测Hep-2细胞增殖活性,Transwell 小室实验检测Hep-2细胞的侵袭力.结果 LSCC组织和癌旁正常组织中OPN阳性表达率分别为75.0％(33/44)、13.6％(6/44),两者相比,P＜0.01;OPN的表达与LSCC颈部淋巴结转移、临床分期、病理组织学分级有关(P均＜0.01).LV-shOPN转染后Hep-2细胞中OPN蛋白表达下降、细胞增殖活性及侵袭力降低(P均＜0.05).结论 OPN在喉癌组织中的表达高于癌旁正常组织,可能参与了喉癌的发生、发展和转移；LV-shOPN能够有效抑制Hep-2细胞的生长和侵袭转移.
