金花茶醇提物对人低分化鼻咽癌CNE-2细胞增殖和周期的影响

         

摘要

目的 探讨金花茶醇提物对人低分化鼻咽癌CNE-2细胞增殖、周期的影响及其机制.方法 将人鼻咽癌CNE-2细胞分为两组,实验组加人终浓度为25、50、100、200、400μg/ml的金花茶醇提物,对照组不加药物,采用MTT法检测两组24、48、72 h增殖抑制率,倒置相差显微镜及荧光显微镜观察48 h后细胞形态的变化,流式细胞仪分析细胞周期变化及凋亡率.结果 金花茶醉提物对CNE一细胞的增殖有抑制作用,且呈时间和剂量效应;金花茶醇提物处理CNE-2细胞48 h后,倒置相差显微镜及荧光显微镜示CNE-2细胞随着浓度的增加发生不同形态改变;流式细胞仪分析显示金花茶醇提物将CNE-2细胞阻滞于G:期.结论 金花茶醇提物能够抑制CNE -2细胞的增殖,其作用机制可能是使CNE-2细胞阻滞于G;期从而抑制CNE一细胞的生长.%Objective To investigate the effect of golden camellias extracts on proliferation, cycle and of poorly differentiated nasopharynx cancer CNE-2 cells and its mechanism. Methods The human nasopharynx cancer CNE-2 cells were divided into two groups,the experimental group was added to the ethanol extract of golden camellias whith the concentration of 25,50, 100,200 and 400 μg/ml; the control group was given no medication. The proliferation inhibition ratio was evaluated by MTT at the time of 24, 48 and 72 hours. Cells morphological changes after 48 hours were observed under the reverse microscope and the fluorescence microscope. Flow cytometry was used to detect phase distribution of cell cycles and apoptosis percentage. Results The ethanol extract of golden camellias inhibited the growth of CNE-2 cells evidently in time and dose -dependent manner. CNE-2 cells occurred various morphological changes with the increasing of concentrations,48 hour after the treatment of extract of golden camellias. Flow cytometry indicated that extract of golden camellias blocked CNE-2 cells in G1 phase. Conclusions The extract of golden camellias can inhibit the proliferation of CNE-2 cells, and its mechanism maybe arrest the CNE-2 cells in G1 phase and thus inhibit the growth of CNE-2 cells.
