转化生长因子-β对人Th17细胞增殖的影响

摘要

Objective To investigate the immunoregulatory function of transforming growth factor -β(TGF-β) on human Th17 cells.Methods Human peripheral blood CD+4 T cells purified by anti-CD4 monoclonal antibody conjugated microbeads from healthy donors were randomly divided into observed group and control group, then both groups were stimulated with 5 μg/ml PHA and 5 ng/ml of recombinant human IL-2 in IMDM medium for 4 days at 37 ℃, with 5％ CO2, the observed group was given human recombined TGF-β( 100 IU/ml) at the same time.The concentrations of IL-17 protein in the supematants were assayed by enzyme-linked immunosorbentassay (ELISA), and quantity of Th17 cells were detected by flow cytometry.Results The level of IL-17 protein in the supematants of CD4+ T cells in the observed group and control group was (6.11 ±9.20), (15.07 ± 12.70)pg/ml, the observed group was lower (P=0.07); percentage of Th17 cells in the observed group and control group was 2.45％ ± 0.62％ and 2.81％ ± 0.44％, the observed group was significantly lower (P ＜ 0.05).Conclusion TGF-β exerts immunoregulatory function on human Thl7 cells by inhibiting its proliferation in vitro.%目的 探讨转化生长因子(TGF)-β对人Th17细胞的免疫调节作用.方法 应用免疫磁珠分离正常人CD+4 T细胞并随机分为观察组和对照组,均置于含5 μg/ml植物凝集素和5 ng/ml人重组IL-2的IMDM培养基中,在此基础上观察组加入100 IU/ml TGF-β,均于37 ℃、5%CO2孵箱中培养4 d.采用ELISA法检测两组细胞上清中IL-17蛋白水平,流式细胞术检测CD+4 T细胞内IL-17比例(Th17细胞数量).结果 观察组和对照组CD+4 T细胞上清中IL-17蛋白水平分别为(6.11 ±9.20 )、(15.07±12.70 )pg/ml,观察组低于对照组(P=0.07);观察组和对照组Th17细胞数量分别为2.45%±0.62%、2.81%±0.44%,观察组明显低于对照组(P＜0.05).结论 TGF-β在体外能通过抑制人Th17细胞增殖而调节其免疫作用.
