p38MAPK介导Ghrelin对TNF-α诱导的人脑微血管内皮细胞MCP-1 mRNA表达的抑制

         

摘要

目的 探讨Ghrelin对TNF-а诱导的人脑微血管内皮细胞(HBMEC)单核细胞趋化蛋白1(MCP-1)的影响及p38MAPK的作用.方法 培养HBMEC,TNF-а组加入不同浓度TNF-а;Ghrelin预处理组先加入Ghrelin预处理1 h后,加入TNF-а.采用RT-PCR法检测MCP-1 mRNA的水平,免疫印迹法检测磷酸化p38(p-p38)的表达.结果 经TNF-а刺激后,MCP-1 mRNA表达明显增强,并呈一定的浓度剂量依赖关系(P<0.05),胞质中的p-p38蛋白表达亦明显增加(P<0.05).Ghrelin预处理可以减少MCP-1 mRNA及胞质中的p-p38蛋白表达(P均<0.05).结论 Ghrelin可能通过抑制p-p38通路抑制TNF-а介导的HBMEC MCP-1 mRNA表达.%Objective To investigate the effect of Ghrelin on monocyte chemoattractant protein-1 ( MCP-1) expression in human brain microvascular endothelial cells ( HBMEC) induced by TNF-α and the effects of p-38 MAPK.Methods After being cultured, different concentration TNF-α were used to treat the HBMEC.In Ghrelin pretreatment group, Ghrelin preprocessing (10 ng/ml) was performed an hour before adding the TNF-α (1.0 ng/ml).The expression of HBMEC MCP-1 mRNA was detected by RT-PCR, and the p-p38 expression was investigated by western blot.Results The expression of MCP-1 mRNA in HBMEC was increased when cells were exposed to different concentration of TNF-α with concentration dependent ( P< 0.05 ) , and the p-p38 expression in cytoplasm increased significantly ( P <0.05) .The expression of MCP-1 mRNA and p-p38 decreased significantly after the Ghrelin preprocessing ( P < 0.05 ) . Conclusion Ghrelin might inhibit the MCP-1 expression in HBMEC via p38 MAPK.
