SLE患者PBMCs细胞凋亡及其相关基因Bax、p53、C-myc蛋白表达的临床意义

         

摘要

目的 探讨系统性红斑狼疮(SLE)外周血单个核细胞(PBMCs)细胞凋亡及其相关基因Bax、p53、C-myc蛋白表达在SLE发病中的作用及机制.方法 应用流式细胞仪(FCM)检测20例活动期SLE患者(SLE组)及20例健康者(对照组)PBMCs细胞凋亡率及凋亡相关基因Bax、p53、C-myc蛋白表达阳性率;免疫比浊法检测血清补体C3水平,并分析SLE组PBMCs细胞凋亡率与C3水平的相关性.结果 SLE组PBMCs细胞凋亡率及Bax、p53、C-myc蛋白表达阳性率均显著高于对照组(P均<0.01);SLE组血清补体C3水平显著低于对照组,且SLE组PBMCs细胞凋亡率与血清补体C3水平呈高度负相关(P均<0.01).结论 Bax、p53、C-myc蛋白表达在SLE发病中具有重要作用,可能机制为诱发PBMCs细胞凋亡失衡及反馈性降低血清补体水平.%Objective To investigate the roles and mechanism of apoptosis and Bax,p53 ,C-myc protein expression of the peripheral blood mononuclear cells(PBMCs) in the genesis of systemic lupus erythematosus (SLE). Methods The apoptosis rate and Bax, p53, C-myc protein expression rate of PBMCs were detected by flow cytometry (FCM) in 20 SLE patients with active disease (SLE group)and 20 healthy controls (control group) ;The C3 serum level was detected quantitatively by turbidimentry, the association between the apoptosis rate of PBMCs and C3 serum level was analysed. Results The apoptosis rate of PBMCs and protein expression rate of Bax, p53, C-myc in SLE group were significantly higher than those in the control group( all P < 0.01 ) ;The C3 serum level in SLE group was significantly lower than that in the control group ( all P <0.01 ) ,there was highly negative correlation between the apoptosis rate of PBMCs and the serum level of C3 in SLE group. Conclusion The expression of Bax,p53 ,C-myc protein play important role in the pathogenesis of SLE ,the possible mechanism maybe inducing apoptosis imbanlance and thereby lowing serum complement level.
