芍药苷预处理对脑缺血再灌注损伤大鼠脑组织细胞凋亡及 CHOP 蛋白表达的影响

         

摘要

目的:观察芍药苷( PA)预处理对脑缺血再灌注损伤( CIR)大鼠脑组织细胞凋亡及CHOP蛋白表达的影响。方法将72只SD雄性大鼠随机分为预处理组、模型组、假手术组各24只,预处理组给予PA 0.2 mg/kg灌胃,假手术组及模型组给予等量PBS灌胃,1次/d,连续3 d。给药后第4天,预处理组、模型组建立CIR模型,假手术组仅暴露各动脉,不做切口与线栓插入。分别于再灌注1(T0)、4(T1)、24(T2) h各取6只大鼠,采用TUNEL法检测脑组织细胞凋亡指数,免疫组化法检测CHOP蛋白表达;于缺血再灌注后24 h各取6只大鼠进行神经功能评分。结果各组同时点脑组织细胞凋亡指数、CHOP蛋白表达比较,模型组>预处理组>假手术组,P均<0.05;预处理组和模型组T2时点脑组织细胞凋亡指数、CHOP蛋白表达均高于同组T0、T1时点,P均<0.05。预处理组神经功能评分为(1.33±0.52)分,模型组(2.50±1.05)分,假手术组为0分;预处理组和模型组的神经功能评分均高于假手术组,P分别<0.05、0.01;预处理组和模型组比较,P<0.01。结论 PA预处理对CIR大鼠脑组织具有保护作用,可能与其降低细胞凋亡及CHOP蛋白表达有关。%Objective To observe the influence of paeoniflorin ( PA) pretreatment to brain cells apoptosis and the ex-pression of CHOP on brain tissue of cerebral ischemia reperfusion injury ( CIR) in rat.Methods 72 male SD rats were randomly divided into 3 groups:pretreatment group, model group, sham operation group, each group of 24 rats.Pretreat-ment group was given PA 0.2 mg/kg by gavage, sham operation group and model group were given equal PBS by gavage once a day for 3 d.The fourth day after the treatment, CIR model were set up in pretreatment group and model group.6 rats in each group were taken out for detecting apoptosis index of brain tissue by TUNEL method, CHOP protein expression by immunohistochemical staining, at 1 h ( T0 ) , 4 h ( T1 ) , 24 h ( T2 ) after reperfusion.In each group, 6 rats were taken out for making neurological severity score at 24 h after reperfusion.Results At the same time point, the brain tissue cell apoptosis index and the CHOP protein expression level, model group>pretreatment group>control group, all P<0.05;the brain tissue apoptosis index and CHOP protein expression at T2 were higher than those in T0 , T1 in pretreatment group and model group, all P<0.05.The score of nerve function in pretreatment group and model group were higher than that in sham operation group, P<0.05 and 0.01 respectively;that in pretreatment group was lower than model group, P<0.01. Conclusion PA pretreatment can protect brain tissue of CIR rat, which maybe correlate with reducing apoptosis, CHOP protein expression and neural function score.
