Objective To study the effects of interleukin 7 ( IL-7) on different subsets of natural killer ( NK) cells in multiple sclerosis ( MS) patients.Methods The peripheral blood mononuclear cells from 10 MS patients were isolated and cultivated, then the subsets of NK cells and the expression of IL-7 receptors alpha (IL-7Rα) were measured.Mononuclear cells were stimulated with 50 ng/mL IL-7 or the same volume of culture media, the percentage of NK cells producing inter-feron gamma ( IFN-γ) and survival rate of NK cells by flow cytometry.Results Phenotype NK cells from MS patients by surface markers included of CD3-CD5+6 , CD3-CD1+6 and CD3-CD5+7 subsets, the mean fluorescence intensity ( MFI) of IL-7Rαexpression were 185.00 ±38.56, 139.30 ±14.30, 137.60 ±23.03, respectively;compared with CD-3 CD+56 NK sub-set, CD-3 CD+16 and CD-3 CD+57 NK subsets had lower IL-7Rαexpression (both P<0.05).Upon IL-7 stimulation, the per-centage of NK cells producing IFN-γincreased by 9.71%±2.59%, and the percentage of survival cells increased by 4.80%±1.88%on CD-3 CD+56 NK subset, which were both higher than those in CD-3 CD+16 and CD-3 CD+57 NK subsets, all P<0.01.Conclusion IL-7 stimulation promotes IFN-γproduction and survival rate of NK cells, and the effects are more prominent on CD3-CD5+6 subset than CD3-CD1+6 and CD3-CD5+7 subsets.%目的：观察白细胞介素7（IL-7）对多发性硬化症（MS）患者不同亚群自然杀伤（NK）细胞的影响。方法抽取10例MS患者的静脉血，提取外周单个核细胞后体外培养，采用流式细胞术检测NK细胞亚群及IL-7受体α（ IL-7Rα）；分别加入50 ng/mL的IL-7或等体积培养液，检测分泌干扰素（IFN-γ）的NK细胞比例及细胞存活率。结果本组MS患者NK细胞分为CD-3 CD＋56、CD-3 CD＋16、CD-3 CD＋573个亚群，其IL-7Rα表达的荧光强度分别为185．0±38．56、139．30±14．30、137．60±23．03；CD-3 CD＋56与CD-3 CD＋16、CD-3 CD＋57亚群比较，P均＜0．05。 IL-7刺激后，CD-3 CD＋56亚群分泌IFN-γ的细胞比例增长了9．71％±2．59％、细胞存活率增长了4．80％±1．88％，均高于CD-3 CD＋16亚群的1．91％±0．93％、2．49％±1．00％和CD-3 CD＋57亚群的2．90％±1．09％、1．87％±0．57％，P均＜0．01。结论 IL-7可以促进各亚群NK细胞分泌IFN-γ并提高其存活率，且对CD-3 CD＋56亚群的影响高于CD-3 CD＋16和CD-3 CD＋57亚群。
