舒心安神膏灌胃对大鼠自主活动的影响及联合戊巴比妥钠腹腔注射对小鼠睡眠和肌肉协调力的影响

         

摘要

Objective To observe the effect of Shuxin Anshen paste on rat′sautonomic activities by intragastric admin-istration and the effect of Shuxin Anshen paste combined with sodium pentobarbital on mouse′s sleep and motor coordination by intraperitoneal injection.Methods Fifty SPF SD rats were randomly divided into groups A, B, C, D and E, each group had 10 rats.Groups A, B, C were separately given 1 mL/100 g Shuxin Anshen paste at the concentration of 0.28 g/mL, 0.84 g/mL and 1.40 g/mL by intragastric administration once a day for 5 days.Group D was given 1 mL/100 g dia-zepam by intragastric administration on the 5th day.Group E was given 1 mL/100 g pure water as the control group.Sixty minutes after the last drug administration, all rats were put in the spontaneous activity box.The times of standing, groo-ming, central grilling and total crossing in 5 minutes were recorded.Fifty NIH mice were randomly assigned into low dosage group, medium dosage group, high dosage group, diazepam group and blank group, each group had 10 mice.Low dosage group, medium dosage group and high dosage group were separately given 0.2 mL/10 g Shuxin Anshen paste at the concen-tration of 0.205, 0.615 and 1.025 g/mL by intragastric administration once a day for 5 days.Diazepam group and control group were separately given 4 mg/kg diazepam and 1 mL/100 g pure water by intragastric administration on the 5th day. Sixty minutes after the last drug administration, each group was separately given pentobarbital sodium 45 ( the up threshold dose) and 40 ( the under threshold dose) mg/kg by intraperitoneal injection.All groups′sleep latency and sleep time was measured and recorded.Fifty NIH mice were randomly assigned into groups 1, 2, 3, 4 and 5.Groups 1, 2 and 3 were giv-en 0.2 mL/10 g Shuxin Anshen paste at the concentration of 0.205, 0.615 and 1.025 g/mL by intragastric administration once a day for 5 day.Groups 4 and 5 were separately given 4 mg/kg diazepam and 0.2 mL/10 g pure water by intragastric administration on the 5th day.Sixty minutes after the last drug administration, each group was put on the rolling-bar device and was observed for 5 minutes.Falling latency and total falling times in 5 minutes were recorded.Results In the auto-nomic activity test, the times of standing and the central grilling of groups A,B,C and D were significantly less than that of group E (all P<0.05).In the sedative and hypnotic experiment using Shuxin Anshen paste and pentobarbital sodium (the up threshold dose) , the sleep latency among low dosage group, medium dosage group, high dosage group, diazepam group and control group was not statistically significant, but the sleep time of these group was significantly higher than that of the control group (all P<0.05).In the experiment of promoting the hypnosis in mice by Shuxin Anshen paste and pentobarbi-tal sodium with the under threshold dose, the sleep latency of low dosage group, medium dosage group, high dosage group, diazepam group and control group was (3.89 ±3.02), (3.20 ±4.32), (2.80 ±2.14), (5.30 ±2.75) and (15 ±0) min.The sleep time of these groups was (36.00 ±4.53), (38.00 ±5.90), (40.29 ±7.36), (36.30 ±10.32) and 0 min.The falling latency of group 4 was significantly less than that of the other groups, and the falling time in 5 minutes of group 4 was significantly higher than that of the other groups (all P<0.05).Conclusions Shuxin Anshen paste can sup-press the rat′s autonomic activity.The synergism of Shuxin Anshen paste and pentobarbital sodium can promote the mouse′s sleep and have little influence on the muscle coordination.%目的:观察舒心安神膏灌胃对大鼠自主活动及联合戊巴比妥钠腹腔注射对小鼠睡眠、肌肉协调运动能力的影响。方法将健康SPF级SD大鼠50只随机分为A、B、C、D、E组,每组10只。 A、B、C组分别予0.28、0.84、1.40 g/mL的舒心安神膏1 mL/100 g灌胃,1次/d,连续5 d。 D组于给药第5天予2 mg/kg安定灌胃。 E组为空白对照组,予1 mL/100 g纯净水灌胃。末次给药60 min后取各组大鼠置于自发活动箱内,记录5 min内直立次数、整毛次数、中央穿格数、总穿格次数;取健康NIH小鼠50只,分为低、中、高剂量组、安定组、空白组各10只。低、中、高剂量组分别予0.205、0.615、1.025 g/mL的舒心安神膏0.2 mL/10 g灌胃,1次/d,连续5 d。安定组和空白组分别于给药第5天予4 mg/kg安定、1 mL/100 g纯净水灌胃。各组分别于末次给药60 min时腹腔注射戊巴比妥钠45(阈上剂量)、40(阈下剂量) mg/kg。观察并计算各组入睡潜伏期和睡眠时间。取健康NIH小鼠50只随机分为1、2、3、4、5组,1、2、3组分别予0.205、0.615、1.025 g/mL的舒心安神膏0.2 mL/10 g灌胃,1次/d,连续5 d。4、5组分别于给药第5 d予4 mg/kg安定、0.2 mL/10 g纯净水灌胃。各组均于末次给药60 min后将各组小鼠置于小鼠转棒仪观察5 min,记录各组小鼠跌落潜伏期和5 min内总跌落次数。结果 A、B、C、D组直立次数和中央穿格数均低于E组(P均<0.05)。舒心安神膏协同阈上剂量戊巴比妥钠促进小鼠催眠实验中,低、中、高剂量组、安定组和空白组入睡潜伏期间比较差异无统计学意义。低、中、高剂量组和安定组的睡眠时间均长于对照组(P均<0.05)。舒心安神膏协同阈下剂量戊巴比妥钠促进小鼠催眠实验中,低、中、高剂量组、安定组和空白组的入睡潜伏期分别是(3.89±3.02)、(3.20±4.32)、(2.80±2.14)、(5.30±2.75)、(15±0) min;睡眠时间分别是(36.00±4.53)、(38.00±5.90)、(40.29±7.36)、(36.30±10.32)、0 min。4组跌落潜伏期少于5组(P<0.05),4组跌落次数明显高于其余各组(P均<0.05)。结论舒心安神膏能抑制大鼠自主活动,可协同戊巴比妥钠促进小鼠睡眠,且对小鼠运动协调能力影响较小。
