葡萄卷叶伴随病毒4和5简并引物和多重PCR检测

摘要

研究建立了葡萄卷叶伴随病毒4和5(Grapevine lea froll associated virus 4 and 5,GLRaV-4 and 5)的简并引物及多重PCR检测方法.比较了单一PCR、多重PCR和简并引物检测方法的灵敏度,并对简并引物的特异性进行了分析.结果表明,采用简并引物和多重PCR对这两种病毒进行检测,其灵敏度与单一PCR相同,且简并引物仅对葡萄卷叶伴随病毒4和5具有特异性.本研究建立的方法能够同时、快速、灵敏地检测上述2种葡萄卷叶病毒,提高了检测效率,降低了检测费用.%The degenerate primer and multiplex PCR methods were established to simultaneously detect Grapevine leafroll-associated virus-i and 5 (GLRaV-4 and 5). The single, multiplex and degenerate primer PCR assays were compared, and the specificity of the degenerate primer pair was analyzed. The results indicated that the sensitivities of these assays were similar, and the degenerate primers were specific to GLRaV-4 and GLRaV-5. Using degenerate primer and multiplex PCR methods to detect GLRaV-4 and 5 could improve efficiency and reduce cost.
