五种昆虫可溶性海藻糖酶活性比较

         

摘要

采用葡萄糖氧化酶法,比较测定了亚洲玉米螟[Ostrinia furnacalis(Guenée)]、黏虫[Mythimna separata (Walker)]、棉铃虫[Helicoverpa armigera(Hübner)]、小菜蛾[Plutella xylostella (Linnaeus)]和甜菜夜蛾[Spodoptera exigua (Hübner)]5种农业害虫的不同生活史阶段及5龄幼虫各组织中可溶性海藻糖酶活性的变化情况.活性测定结果表明:5种昆虫卵中海藻糖酶的活性最低,其次为蛹和初孵幼虫,取食阶段海藻糖酶的活性比较高,随着幼虫龄期的增加,海藻糖酶的活性也随着升高.亚洲玉米螟、黏虫、棉铃虫和甜菜夜蛾不同组织部位海藻糖酶的活性研究表明,亚洲玉米螟和棉铃虫中肠中海藻糖酶的活性最高,分别为316.17和39.35 mU/mg,而甜菜夜蛾体壁中海藻糖酶活性最高,达到139.99 mU/mg.黏虫脂肪体中海藻糖酶活性最高,为11.19 mU/mg.不同昆虫间海藻糖酶的活性差异较大,玉米螟和甜菜夜蛾各组织的海藻糖酶活性较高,而黏虫各组织部位海藻糖酶的活性很低.%The activities of the soluble trehalase in five agricultural pest species,namely Ostrinia furnacalis (Guenée),Mythimna separata (Walker),Helicoverpa armigera (Hübner),Plutella xylostella (Linnaeus)and Spodoptera exigua (Hübner),were determined in different tissues and at different developmental stages by using the method of glucose oxidase.The results showed that the activity of trehalase in eggs was the lowest,followed by those in pupae and initial hatched larvae.The trehalase activity was found relatively high during the feeding stage,which was increased with the larval stages.The highest trehalase activities were found in midgut of O.furnacalis (316.17 mU/mg.) and H.armigera (39.35 mU/mg),while in body wall of S.exigua and in fat body of M.separata,the highest trehalase activity were 139.99 and 11.19 mU/mg,respectively.The trehalase activity also showed significant difference among different insects.The trehalase activity was relatively high in different tissues of O.furnacalis and S.exigua,but low in tissues of M.separata.
