维吾尔族宫颈癌组织中蛋白酶激活受体3(Par3)与上皮-间质转化相关蛋白的表达及其临床意义

         

摘要

Abstact:Objective To investigate the expression levels and correlation of polar protein E-cadherin and ep-ithelial marker Par3,E-cadherin,N-cadherin andβ-catenin,Smad1 and vimentin in cervical squamous cell carcinoma (CSCC),cervical intraepithelial neoplasia (CIN).Methods CSCC,CIN patients and normal cervical paraffin embedded tissue specimens of 189 cases were obtained to detect the expression of Par3, E-cadherin,N-cadherin,β-catenin,Smad1,vimentin by immunohistochemistry.Results Par3,E-cadherin, N-cadherin,β-catenin,Smad1 and vimentin showed different degrees of positive expression in cervicitis, CIN and CSCC tissues.The positive expression rates of Par3,E-cadherin,β-catenin,N-cadherin,Smad1 and vimentin in normal cervical tissues were 85.4%,53.7%,19.5%,53.7%,22% and 26.8%,respectively. The positive expression rates of CIN were 50%,31.3%,34.3%,9.3%,56.3% and 43.8%,respectively. In cervical cancer,the positive expression rates were 17.2%,19.5%,77.6%,8.6%,76.7% and 87.1%, respectively.And with the increase of cervical lesions,Par3,E-cadherin and β-catenin expressions were down regulated (P <0.05),while increased expression of N-cadherin,Smad1 and vimentin (P <0.05). The expression of PARD3,E-cadherin,N-cadherin,β-catenin and Smad1 were related to the FIGO stage, differentiation degree and lymph node metastasis of cervical cancer.The expression of vimentin was related to the degree of differentiation,differentiation and lymph node metastasis of invasive cervical cancer,and the expression difference was statistically significant (P <0.05).Conclusion With the progress of cervical lesions Par3 expression and epithelial markers E-cadherin,andβ-catenin expression showed positive corre-lation,and negatively correlated with mesenchymal markers N-cadherin,Smad1,vimentin expression in cervical cancer,suggesting that low expression of Par3 protein may be involved in the process of cervical epithelial mesenchymal transition.%目的:研究维吾尔族宫颈炎、宫颈上皮内瘤变和宫颈癌组织中极性蛋白 Par3与上皮型钙黏蛋白(E-cadherin)、β-连环蛋白(β-catenin)、神经性钙黏蛋白(N-cadherin)、转录因子 Smad1、波形蛋白(Vimentin)的表达及其临床病理学意义。方法收集维吾尔族妇女宫颈鳞癌(cervical squamous cell carcinoma,CSCC)、宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)患者和正常宫颈石蜡包埋组织标本189例,采用免疫组织化学的方法分别检测 Par3与 E-cadherin、N-cadherin、β-catenin、Smad1及 vimentin 的表达情况。结果Par3、E-cadherin、N-cad-herin、β-catenin、Smad1及 vimentin 在正常宫颈组织、CIN 和 CSCC 组织中呈不同程度阳性表达。正常宫颈组织中Par3、E-cadherin、N-cadherin、β-catenin、Smad1及 vimentin 阳性表达率分别为85.4%、53.7%、19.5%、53.7%、22.0%、26.8%。在 CIN 组织中阳性表达率分别为50.0%、31.3%、34.3%、9.3%、56.3%、43.8%。在宫颈癌中阳性表达率分别为17.2%、6.8%、77.6%、8.6%、76.7%、87.1%。随着宫颈病变的加重,Par3、E-cadherin、β-catenin 的表达显著下调(P <0.05)。 N-cadherin、Smad1及 vimentin 表达显著上调(P <0.05)。 Par3及 E-cadherin、N-cadherin、β-catenin、Smad1蛋白的表达均与宫颈癌 FIGO 分期、分化程度及淋巴结转移有关(P <0.05)。而Vimentin的表达则仅与浸润性宫颈癌组织分级、分化程度及淋巴结转移有关(P <0.05)。结论随着宫颈病变的进展,宫颈癌组织中 Par3蛋白低表达可能参与了宫颈上皮间充质转变的过程。
