冠心病猝死心肌中泛素活化酶UBE1、端粒酶的表达及意义

         

摘要

目的 探讨泛素活化酶UBE1、端粒酶在冠心病猝死心肌中的表达及意义.方法 收集冠心病心源性猝死心肌28例、冠心病非心源性猝死心肌10例,10例正常心肌作为对照,应用免疫组化(EnVision二步法)技术及Image-Pro Plus 6.0图像分析软件检测各组UBE1、端粒酶表达的累积光密度(IOD).结果 Image-Pro Plus 6.0图像分析软件显示UBE1在冠心病心源性猝死组、冠心病非心源性猝死组及正常对照组中累积光密度(IOD)的中位数(四分位数间距)依次为1 182.77(952.82-1 567.32)、892.69(603.38-1 146.79)及833.52(752.74-873.54),端粒酶在以上三组中IOD的中位数(四分位数间距)依次为12 544.9(11 128.29-13 502.25)、6 368.07(3 648.35-12 485.16)及8 658.08(8 058.23-10 066.13).UBE1、端粒酶在冠心病心源性猝死组中的累积光密度(IOD)显著高于冠心病非心源性猝死组及正常对照组,组间差异有统计学意义(P<0.05).结论 UBE1和端粒酶在冠心病心源性猝死心肌中呈过表达,UBE1和端粒酶有望成为心肌早期损伤的标志物.%Objective To investigate the expression and significance of ubiquitin activating enzyme ( UBE1) and telomerase in the myocardium from sudden coronary death.Methods Myocardia from 28 cases of cardiac sudden coronary death , 10 cases of non-cardiac sudden coronary death and 10 cases of normal controls were collected .Immunohistochemistry (EnVision two-step method) and Image-Pro Plus 6.0 image analysis software were employed to detect the integrated optical density (IOD) of UBE1 and telomerase expression.Results The median (interquartile range) IOD of UBE1 was 1 182.77(952.82-1 567.32 ) ,892.69 (603.38 -1 146.79) and 833.52(752.74 -87354 )in cardiac sudden coronary death group , non-cardiac sudden coronary death group and control group , respectively ; the median (interquartile range ) IOD of telomerase in the three groups was 12 544.9 ( 11 128.29 - 13 502.25 ), 6 368.07 (3 648.35 -12 485.16) and 8 658.08(8 058.23 -10 066.13) ,respectively.IODs of UBE1 and telomerase in cardiac sudden coronary death group were significantly higher than those in non -cardiac sudden coronary death group and normal control group ( P < 0.05 ).rnConclusion UBE1 and telomerase are overexpressed in the myocardia in sudden coronary death patients , and may be markers for diagnosing the early myocardial injury.
