转基因741杨杂种胚培养与植株再生

         

摘要

We studied the immature embryo induced germination , adventitious bud regeneration , bud subculture and rooting cul-ture by using immature embryos of transgenic 741 poplar hybrids as explants, then obtained the regenerated plants.The immature embryos get a highest induced germination rate of 76.5%when adding 6-BA (0.1 mg/L), 0.01 mg NAA and sucrose (30 g/L) in medium MS.The appropriate period to take embryo is from embryo age of 20-27 days until fully ma-ture.There are obvious differences of immature embryo induced germination rate in different hybrid combinations, and the transgenic lines pb1×84K have the highest induced germination rate of 66.1%.The ingredients of adventitious buds sub-culture medium are 6-BA (0.2-1.0 mg/L) and IBA (0.05-0.3 mg/L) in medium MS.The ingredients of root medium are IBA (0.2-0.5 mg/L) and sucrose (1.5%) in medium 1/2MS, and the rooting rate is more than 95%.%以转基因741杨杂种未成熟胚为外植体,对未成熟胚诱导萌发与不定芽再生,以及芽的继代与生根培养进行了研究并获得再生植株。结果表明,最适宜未成熟胚诱导萌发的培养基为MS+6-BA0.1 mg · L-1+NAA0.01 mg· L-1+3%蔗糖,诱导萌发率最高可达76.5%;未成熟胚离体培养最适取胚时间为胚龄20~27 d;不同杂交组合未成熟胚诱导萌发率存在明显差异,最高的杂交组合为转基因株系pb1×84K,诱导萌发率为66.1%。不定芽继代培养基为MS+6-BA(0.2~1.0 mg/L)+IBA(0.05~0.3 mg/L);生根培养基为1/2MS+(0.2~0.5 mg/L) IBA+1.5%蔗糖,生根率在95%以上。
