甲基化干预在肿瘤治疗中的研究进展

摘要

表观遗传学在肿瘤发生发展中具有重要的作用,其中DNA甲基化被认为是肿瘤形成的重要机制之一,抑癌基因通常因高度甲基化而失活.由于DNA的甲基化状态可被逆转,改善其甲基化状态就有可能使基因恢复表达,这为恶性肿瘤的治疗提供了新的思路.本文就DNA甲基化干预肿瘤治疗情况作一综述,说明了DNA甲基化与肿瘤的关系,及DNA甲基化干预的原理,探讨利用5-Aza-CdR进行甲基化干预治疗肿瘤的可行性.%Epigenetic inheritance plays an important role in the appearance and development of cancer.One kind of Epigenetic inheritance called DNA methylation has been thought to be one of mechanisms.Tumor suppressor genes usually lose their function because of being highly methylated.DNA methylation state can be reversed, so demethylation might make tumor suppressor genes expressing again,which provides new ideas for the treatment of malignant tumor.This paper based on the actuality of demethylation in cancer treatment,illustrates the relationship between DNA methylation and cancer, reveals the mechanism of DNA demethylation,discusses the feasibility of demethylation by using 5 - Aza - CdR to treat cancer. .
