槲皮素对人结肠癌 SW480 细胞体外侵袭能力的影响

摘要

目的:研究槲皮素对结肠癌SW480细胞的体外侵袭能力的影响,并探讨其可能的作用机制.方法:结肠癌SW480细胞经槲皮素处理后用Transwell小室的方法检测其体外黏附、侵袭和运动能力.明胶酶谱分析法检测SW480细胞分泌的基质金属蛋白酶活性.结果:结肠癌SW480细胞经槲皮素处理后,体外侵袭、运动能力呈剂量依赖性下降,SW480细胞MMP-2及MMP-9的分泌下降(P＜0.05).结论:槲皮素在体外剂量依赖性地抑制结肠癌SW480细胞的转移能力,这可能与槲皮素抑制MMP-2及MMP-9的分泌有关.%Objective:To observe the effect on the invasion of human colon cancer SW480 cell line in vitro induced by quercetin. Methods: Human colon cancer cells SW480 were treated with different doses of quercetin, and the invasion of SW480 was determined by Transwell chamber, the expressions of MMP - 2 and MMP - 9 were detected by gelatin zymography. Results; Transmembrane cell and the expressions of MMP -2 and MMP - 9 of quercetin group decreased in dose - dependent manners (P <0. 05 ). Conclusion; Quercetin can inhibit invasion of human colon cancer cells, and decreasing the expression of MMP - 2 and MMP - 9 may play an important role in its mechanism.
