目的:探讨miR-17-5p在膀胱癌细胞中的表达及其对膀胱癌T24细胞生长的影响.方法:Real-time qRT-PCR(quantitative reverse transcription PCR)方法检测miR-17-5p在膀胱癌5637、T24、SW780细胞系中的表达.化学合成anti-miR-17-5p抑制剂,瞬时转染T24细胞.应用MTT实验和流式细胞术分别检测T24细胞低表达miR-17-5p对细胞增殖、凋亡的影响.结果:miR-17-5p在3种膀胱癌细胞系中均具有高表达特性,体外抑制T24细胞miR-17-5p表达能降低其增殖活力和抗凋亡能力.结论:miR-17-5p具有促进细胞增殖、抑制细胞凋亡的作用,miR-17-5p高表达可能参与了膀胱癌发生发展.%Objective:To investigate the expression and biological function of miR-17-5p in bladder cancer cells.Methods:The expression of miR-17-5p in bladder cancer cell lines-5637,T24 and SW780 was determined by real-time quantitative reverse transcription PCR(qRT-PCR).Anti-miR-17-5p inhibitor was trans-iently transfected into T24 cells.Proliferation and apoptosis of the cells were evaluated using MTT assays and flow cytometry.Results:miR-17-5p was up-regulated in 5637,T24 and SW780 cells compared with that in HUC cells.Inhibition of miR-17-5p in T24 cells contributes to decrease of cell proliferation and anti-apoptosis.Conclusion:miR-17-5p can promote cell proliferation and inhibit apoptosis,and the aberrant increased expression of miR-17 -5p may contribute to bladder carcinogenesis.
