沉默食管鳞癌细胞PLCE 1基因表达后cyclin D1和caspase-3的变化

         

摘要

目的:通过siRNA沉默PLCE1基因表达,研究PLCE1对食管鳞癌细胞增殖周期和凋亡相关调控蛋白的影响,对其致癌机制进行探讨。方法:PLCE1特异性siRNA转染食管癌细胞,荧光显微镜观察转染效率;半定量RT-PCR法检测转染后PLCE1沉默效果;Western blot检测沉默后cyclin D1和caspase-3的表达水平。结果:siRNA转染后PLCE1表达明显减少(P=0.000);cyclin D1水平降低(P=0.03)、caspase-3表达升高(P=0.027)。结论:PLCE1基因可能作为癌基因参与了食管癌的发生和发展,其机制可能和PLCE1与某些调控细胞周期和凋亡的蛋白表达有关。%Objective:To investigate the effects of PLCE1 (phospholipase Cε1 )gene silencing by siRNA (small interfering RNA)interference on certain protein which control cell cycle and apoptosis of esophageal squamous carci-noma cells(ESCC),to identify the role of PLCE1 in esophageal squamous cancer.Methods:ESCC 9706 were trans-fected with the specific siRNA of PLCE1.Fluorescence microscopy was used to examine transfection efficiency,the re-sult of PLCE1 silencing was examined by reverse transcription RT-PCR.Expression of cyclin D1 and caspase -3 was detected by Western blot.Results:After transfection,mRNA expression of PLCE1 in ESCC 9706 was significantly reduced (P=0.000).The expression of cell cycle and apoptosis related proteins-such as cyclinD1 and caspase-3, showed that the down-regulation of PLCE1 could inhibit the expression of the former(P=0.03)while promote the other (P=0.027).Conclusion:The PLCE1 gene may be an oncogene and play an important role in esophageal car-cinogenesis through regulating certain protein which control cell cycle and apoptosis.
