β-榄香烯联合顺铂对乳腺癌 MCF -7细胞增殖和凋亡的影响

摘要

目的：探讨β-榄香烯联合顺铂对乳腺癌 MCF -7细胞生长的影响。方法：体外培养乳腺癌 MCF -7细胞，将β-榄香烯（浓度梯度为25、50、100、150、200μg/ ml），单独作用于乳腺癌 MCF -7细胞，加药24h、48h后用噻唑蓝（MTT 法）法检测细胞增殖情况。用流式细胞术检测用药24h 后对 MCF -7细胞凋亡和细胞增殖周期的影响，选取合适的药物浓度（β-榄香烯125μg/ ml），与顺铂（3μg/ ml）进行联合用药。加药24h、48h用 MTT 法检测细胞增殖情况，用流式细胞术检测24h 后药物组对 MCF -7细胞凋亡和细胞增殖周期的影响。结果：MTT 法结果显示β-榄香烯单独用药24h、48h 后，与对照组相比，实验组乳腺癌 MCF -7细胞的抑制率高于对照组（P ﹤0.05），并且在一定程度上呈浓度和时间依赖性。联合用药时，细胞的抑制率和凋亡率要显著高于单独用药（P ﹤0.01）。与对照组相比，榄香烯能够使 MCF -7细胞产生明显的 G0/ G1期阻滞（P%Objective:To investigate the effect of β - Elemene combined with DDP on the Proliferation and aPoP-tosis of MCF - 7 cell. Results:Cell Proliferation was dramatically inhibited by β - Elemene injection(25,50,100,150,200μg/ ml)at 24h,48h(P ﹤ 0. 05). In addition,cell Proliferation was dramatically inhib-ited by injection β - Elemene combined with DDP. Combination theraPy,aPoPtosis rate and inhibition rate of Prolifera-tion in MCF - 7 cells treated by β - Elemene combined with DDP was higher than the monotheraPy(P ﹤ 0. 01). Cell- cycle was arrested in G0 / G1 Phase by elemene comPared to uninfectious cells in the same time scale. Conclusion:β - Elemene and DDP combined with β - Elemene can inhibit the Proliferation,accelerate the aPoPtosis in MCF - 7 cells. DDP combined with β - Elemene have synergistic effect on the aPoPtosis in MCF - 7 cells.
