槲皮素抑制X线照射后宫颈癌Hela细胞侵袭性的作用

         

摘要

Objective:To investigate the effects of radiation and quercetin on the invasion of cervical cancer Hela cell and the mechanism. Methods:MTT assay was performed to evaluate the cytotoxicity of quercetin on the Hela cel1s. The effect of radiation and different concentration of quercetin on invasive ability of Hela cells was observed with transwel assay. The protein expression of VEGF and MMP-9 in Hela cells was detected by Western blotting. Re-sults:MTT result showed that the inhibition of quercetin on the cells was in dose-dependent and time-dependent( P﹤0. 05). Compared with the blanK group,radiation transiently increased the cell invasion ability(P﹤0. 01),querce-tin could inhibitory effect on the invasion of Hela(P﹤0. 01),quercetin and ray processing cell,still had obvious in-hibitory effect of invasion(P﹤0. 01). The levels of protein of VEGF and MMP-9 increased in a certain time by ra-diation alone(P﹥0. 05). The levels of protein of VEGF and MMP-9 were decreased by quercetin and radiation combined(P﹤0. 05). Conclusion:Hela cells enhanced invasive potential in a short time after radiation. Quercetin can inhibit the migration and invasion of Hela cells in vitro. The mechanism may be related to the expression of VEGF,MMP- 9.%目的:研究放射线及槲皮素对人宫颈癌Hela细胞侵袭性变化的影响,并初步探讨VEGF、MMP-9表达对其影响。方法:MTT法检测不同浓度的槲皮素对宫颈癌Hela细胞增殖的影响;Transwell体外侵袭实验分别观察放射线及不同浓度槲皮素作用后24h细胞的侵袭能力的改变。Western blot法检测不同浓度槲皮素、放射线单独及联合应用后Hela细胞中VEGF、MMP-9蛋白的表达。结果:槲皮素对Hela细胞具有增殖抑制作用,并呈浓度和时间依赖性( P﹤0.05),24小时的IC50值约为120μmol/L。与空白组比较,X线照射24h后细胞侵袭能力增加(P﹤0.01),槲皮素对HeLa细胞的侵袭能力具有抑制作用(P﹤0.01),槲皮素与X线共同处理细胞后,仍具有明显的抑制侵袭能力( P﹤0.01),组间差异具有统计学意义。X线单独作用后VEGF、MMP-9蛋白表达在一定时间内有增加的趋势( P﹥0.05),槲皮素、以及联合X线组VEGF、MMP-9蛋白表达降低( P﹤0.05)。结论:X线照射后宫颈癌Hela癌细胞侵袭潜能短暂增强;槲皮素能抑制X线照射后宫颈癌细胞的侵袭能力。机制可能与槲皮素能下调VEGF、MMP-9的表达有关。
