结肠癌组织中EZH2和PTEN基因表达与临床病理特征的相关性分析

摘要

目的：检测 EZH2和PTEN基因在结肠癌中的表达，探讨两基因表达与结肠癌患者临床病理因素的关系，同时探讨结肠癌组织中两基因表达之间的关系。方法：选择2011年4月-2013年8月结肠癌标本66例，另取同期20例活检正常结肠黏膜组织标本作为对照，应用免疫组织化学方法分别检测EZH2和PTEN基因在结肠癌及正常结肠黏膜组织中的表达情况。结果：EZH2、PTEN在结肠癌组织中阳性表达率分别为71.21％（47/66）、51.52％（34/66），正常结肠黏膜组织中无EZH2阳性表达，而PTEN阳性表达率为100.00％（20/20）。结肠癌中EZH2、PTEN表达率与正常结肠黏膜组织比较，差异有统计学意义（ P<0.05）。EZH2和PTEN基因的表达与结肠癌患者的Duke分期和有无淋巴结转移有关，而与性别、年龄和组织学分型无关。结肠癌组织中EZH2和PTEN的表达呈负相关（r＝-0.349，P＝0.004）。结论：EZH2表达上调及PTEN表达缺失与结肠癌的发生、发展、浸润、转移有关，检测两指标有助于对结肠癌患者分期的评估及预后的判断。%Objective:To detect the expression of EZH2 and PTEN protein in colon cancer tissue,explore the rela-tionship between the two gene expression and clinicopathological features and the relationship between them. Meth-ods:Immunohistochemical staining was used to detect the EZH2 and PTEN protein expression in the tissues of 66 co-lon cancer from April 2011 to August 2013 in our hospital and 20 normal colonic mucosa at the same time. Results:The expression rates of EZH2 and PTEN in colon cancer rates were 71. 21%(47/66),51. 52%(34/66)respective-ly. There was no EZH2 expression,and the PTEN expression rate was 100. 00%(20/20)in the normal colonic muco-sa tissues. There were significant differences of the expressions of EZH2 and PTEN between the colon cancer tissue and normal colonic mucosa(P<0. 05). The expression of EZH2 and PTEN were closely correlated with Duke stage and lymph node metastasis. Expressions of EZH2 and PTEN had a significant negative corelation(r= -0. 349,P=0. 004). Conclusion:The expression of EZH2 and PTEN may play an important role in development,invasion and metastasis in colon cancer. Detection of two indicators is helpful in the diagnosis and treatment of colon cancer and as-sessment of its biological behavior.
