癌基因 MSP58在骨肉瘤组织中的表达及其对骨肉瘤细胞增殖的影响

摘要

Objective:To explore the expression of MSP58 in osteosarcoma tissues and its effects on the prolifera-tion of osteosarcoma cell.Methods:Real -time PCR and Western blot were used to detect MSP58 mRNA(n =15) and protein(n =7)levelin paired osteosarcoma and peritumor tissues.MSP58 targeting siRNA was transfected into U2OS osteosarcoma cell.Effect of MSP58 on the proliferation and cell cycle of osteosarcoma cell was analyzed by MTT assay and flow cytometry.Results:Upregulation of MSP58 expression was found in osteosarcoma tissues,compared with peritumor tissues(P <0.05).Proliferation of U2OS cells was inhibited dramatically in U2OS cells with transfec-tion of MSP58 targeting siRNA.Meanwhile,downregulation of MAP58 by specific siRNA induces increase G1 phase U2OS cells,while G2 /Mand S phase cells decreased significantly.Conclusion:The expression of MSP58 increases in osteosarcoma tissues,and downregulation of MSP58 inhibits proliferation of U2OS osteosarcoma cell.%目的：研究 MSP58基因在骨肉瘤组织中的表达，探讨干涉 MSP58基因对体外培养的 U2OS 骨肉瘤细胞增殖的影响。方法：利用 Real －time PCR（n ＝15）和 Western blot（n ＝7）方法分别检测新鲜骨肉瘤及癌旁组织中 MSP58 mRNA 和蛋白的表达情况，并以癌旁组织作为对照。用癌基因 MSP58特异的 siRNA 转染 U2OS骨肉瘤细胞，而后采用 MTT 法检测细胞增殖情况，流式细胞术检测细胞周期。结果：与癌旁组织相比，MSP58n　　基因 mRNA 和蛋白水平在骨肉瘤组织中的表达明显升高（P ＜0．05）。干涉 MSP58基因表达明显抑制 U2OS骨肉瘤细胞增殖，同时 G1期细胞明显增多，G2／M期和 S 期细胞明显减少（P ＜0．05）。结论：MSP58基因在骨肉瘤肿瘤组织中过度表达，下调 MSP58表达可抑制骨肉瘤细胞增殖。
