Survivin 与乳腺癌雌激素受体信号通路相关基因的相关性

Objective:To investigate the correlation between Survivin and estrogen receptor signal pathway related genes in breast cancer.Methods:Expression of Survivin in 10 breast cancer tissues were detected by Real -time PCR method,3 cases with the highest expression were selected as the high expression group,and 3 cases with the lowest ex-pression were selected as the low expression group.Different expression of the related genes between the two groups was detected by using human breast cancer estrogen receptor signaling pathway PCR arrays.Results:The expression of Survivin in the two groups was significantly different(P <0.05).The results of detection with the estrogen receptor signal pathway PCR arrays showed that the ER expression in Survivin low expression group was lower than that in Sur-vivin high expression group,but the difference was not significant(P >0.05 ),and the expression of CDKN1B, ERBB2,NME1,TOP2A was significantly lower in Survivin low expression group than that in Survivin high expression group,the differences were significant(P <0.05).Conclusion:In breast cancer,there is positive correlation between Survivin and CDKN1B,NME1,ERBB2,TOP2A in mRNA level,and there is a positive correlation tendency between Survivin and ER expression,which suggested that there may be a correlation between Survivin and ER signal pathway, Survivin is of potential value in breast cancer endocrine therapy.%目的：探讨乳腺癌中 Survivin 与雌激素受体（estrogen receptor，ER）信号通路相关基因的相关性。方法：采用 Real －time PCR 技术检测10例乳腺癌组织中 Survivin 的含量，选择含量最高的3例作为高表达组，含量最低的3例作为低表达组；采用人乳腺癌雌激素受体信号通路 PCR 芯片检测两组标本中相关基因表达的差异。结果：两组标本中 Survivin 的含量差异有统计学意义（P ＜0．05）。雌激素受体信号通路相关基因PCR 芯片检测结果表明，Survivin 低表达组的 ER 含量较高表达组降低，但差异无统计学意义（P ＞0．05）；Sur-vivin 低表达组的 CDKN1B、NME1、ERBB2、TOP2A 含量较高表达组显著降低，差异有统计学意义（P ＜0．05）。结论：在乳腺癌中，Survivin 与 CDKN1B、NME1、ERBB2、TOP2A 的表达在 mRNA 水平上呈正相关，与 ER 的表达存在着正相关的趋势，表明 Survivin 与雌激素受体信号通路之间存在着一定的相关性。Survivin 在乳腺癌的内分泌治疗中可能具有潜在的重要意义。
