Nav1.7与L-型钙离子通道在宫颈癌中的表达及临床意义

         

摘要

目的:探讨钙离子通道1.7亚型(Nav 1.7)与 L -型钙离子通道在子宫颈癌中的表达及临床意义。方法:采用逆转录-聚合酶链反应(RT -PCR)方法检测60例宫颈癌组织、10例经放化疗前后的宫颈癌组织、10例正常宫颈组织中的 Nav 1.7与 L -型钙离子通道的表达。结果:Nav 1.7在宫颈癌中平均值为1.6403,在正常宫颈组织中不表达,两者相比有显著性差异(P <0.01)。Nav 1.7 mRNA 在高分化癌、中分化癌及低分化癌中的表达分别为0.9601、0.9918及1.9014,低分化组 Nav 1.7 mRNA 表达高于高、中分化组,差异有统计学意义(P <0.05)。Nav 1.7 mRNA 在宫颈癌Ⅰ-Ⅱ期的表达为0.9867,在宫颈癌Ⅲ-Ⅳ期中的表达为1.8993,差异有统计学意义(P <0.05),Nav 1.7 mRNA 在放化疗前的宫颈癌组织中表达为1.4010与放化疗后的0.3001相比,有显著性差异(P <0.01)。L -型钙离子通道α1 C 亚单位 mRNA(L -Ca2+α1 C mRNA)在正常宫颈组织中与宫颈癌组织中的灰度值的平均值为0.0914及0.3573,差异有统计学意义(P <0.05)。L -Ca2+α1 C mRNA 在高分化癌、中分化癌及低分化癌中的表达分别为0.2912、0.3043及0.4935,差异有统计学意义(P <0.05)。L -Ca2+α1 C mRNA 在宫颈癌Ⅰ-Ⅱ期中的表达为0.4754,在宫颈癌Ⅲ-Ⅳ期中的表达为0.8337,差异有统计学意义(P <0.01)。L -Ca2+α1 C mRNA 在放化疗前的宫颈癌组织中的表达为0.5316与放化疗后的0.1024相比,有显著性差异(P <0.01)。结论:Nav 1.7与 L -型钙离子通道在宫颈癌中高表达,与宫颈癌的不良预后有关。放化疗可降低 Nav 1.7与 L -型钙离子通道基因的表达,与疗效相关。%Objective:To explore expression and clinical significance of Nav 1.7 and L -type calcium channel subunit α1 C in cervical cancer.Methods:Using reverse transcription -polymerase chain reaction (RT -PCR)meth-ods to detect expression of Nav 1.7 and L -type calcium channel subunit α1 C in 60 cases of cervical cancer tissue,10 cases of cervical cancer who before and after the radiation and chemotherapy group and 10 cases of normal cervical tissue.Results:The ratio of gray level of Nav 1.7 mRNA was 1.640 3 in cervical cancer,but no expression in normal cervix,in which there was significantly difference(P <0.05).The midterm expression of Nav 1.7 mRNA in high dif-ferentiation of carcinoma,differentiation of carcinoma and poorly differentiated carcinoma were 0.960 1,0.991 8 and 1.901 4,respectively,there was significantly difference(P <0.05).The midterm expression of Nav 1.7 mRNA in cervical cancer of stageⅠ -Ⅱwas 0.986 7,while in cervical cancer of stage Ⅲ -Ⅳ was 1.899 3,and the difference was statistically significant (P <0.01).Nav 1.7 was higher before radiotherapy and chemotherapy (1.4010)than that after radiochemotherapy (0.300 1)and the difference was statistically significant(P <0.01).L -type calcium chan-nel subunit α1 C mRNA in normal cervical tissues and cervical cancer tissue of the grey value of ratio were 0.091 4 and 0.357 3,respectively,there was significantly difference(P <0.05).The midterm expression of L -type calciu mchannel subunit α1 C mRNA in high differentiation of carcinoma,differentiation of carcinoma and poorly differentia-ted carcinoma were 0.291 2,0.304 5 and 0.493 5 respectively,there was significantly difference(P <0.05).The midterm expression of L -type calcium channel subunit α1C mRNA in cervical cancer of stageⅠ -Ⅱ was 0.475 4, while in cervical cancer of stage Ⅲ -Ⅳ was 0.833 7,and the difference was statistically significant (P <0.01).L -type calcium channel subunit α1 C mRNA was higher before radiotherapy and chemotherapy (0.531 6)than that after radiochemotherapy(0.102 4)and the difference was statistically significant (P <0.01).Conclusion:L -type calci-um channel subunit α1 C and Nav 1.7 gene are involved in malignant cervical cancer cell proliferation.L -type calciu mchannel subunit α1 C and Nav 1.7 gene are associated with the malignant progression,clinical staging and pathologic stage of cervical cancer.Cervical cancer radiation and chemotherapy can decrease the L -type calcium channel sub-unit α1 C and Nav 1.7 gene expression.
