HMGN5基因在肺癌中的表达及其意义

摘要

目的:探究高迁移率族核小体结核域5(high mobility group nucleosome 5,HMGN5)基因在肺癌细胞中的表达和意义.方法:选取2015年1月至2017年1月于我院确诊肺癌的患者60例,收集标本作为观察组;另取正常肺组织20例作为对照组.采用实时荧光定量聚合酶链式反应(polymerase chain reaction,PCR)技术检测组织中HMGN5基因的mRNA表达水平,通过免疫组化技术检测组织中HMGN5蛋白表达水平,比较肺癌和正常肺组织中上述指标的表达差异及影响表达的因素.结果:观察组HMGN5基因的mRNA表达水平高于对照组,差异有统计学意义(P＜0.05).观察组HMGN5蛋白的表达阳性率高于对照组,差异有统计学意义(P＜0.05).HMGN5蛋白在肺癌中的表达与年龄和性别无关,而同分化程度和临床病理分期(tumor nodemetastasis,TNM)有关,分化程度低、TNM分期高的标本HMGN5蛋白的阳性表达程度高于分化程度高、TNM分期低的标本,差异有统计学意义(P＜0.05).结论:HMGN5基因在肺癌组织中表达显著增加,表达程度与分化程度和TNM分期有关.%Objective:To explore the expression and significance of high mobility group nucleosome 5 (HMGN5) in lung cancer cell.Methods:60 cases of patients with lung cancer treated in our hospital from January 2015 to January 2017 were selected as observation group.20 cases of normal lung tissues were selected as control group.The expression differences and influencing factors of HMGN5 were compared between two groups through polymerase chain reaction and immunohistochemical technique.Results:The expression level of HMGN5 gene in observation group was higher than that in control group(P ＜ 0.05).The positive expression rate of HMGN5 in observation group was higher than that in control group(P ＜ 0.05).The expression degree of HMGN5 was related to differentiation degree and clinical pathological stage.The expression degree of HMGN5 in low degree of differentiation and high clinical stage was higher than that in high degree of differentiation and low clinical stage (P ＜ 0.05).Conclusion:The expressions of HMGN5 are significantly increased in colon carcinoma tissue.They are related to degree of differentiation and TNM stage.
