目的:研究核富含丰富的转录本1_2 (nuclear-enriched abundant transcript 1_2,NEAT1_2)在人子宫颈癌中的表达及其与临床病理特征的关系.方法:采用实时定量PCR(Real time-PCR)法检测NEAT1和NEAT2在10例慢性宫颈炎、10例高级别宫颈上皮内瘤样病变(CINⅡ-Ⅲ)和60例子宫颈癌中的表达,并分析NEAT1和NEAT2表达与子宫颈癌患者临床病理特征的相关性.结果:慢性宫颈炎组织NEAT1和NEAT2的相对表达量为0.34±0.10和0.29±0.06;CINⅡ-Ⅲ的宫颈上皮内瘤变组织NEAT1和NEAT2的相对表达量为1.18±0.32和1.37 ±0.44;子宫颈鳞癌组织NEAT1和NEAT2的相对表达量为6.25±1.58和5.96±1.47,子宫颈腺癌组织NEAT1和NEAT2的相对表达量为5.03±1.12和4.96±1.03.NEAT1和NEAT2在子宫颈癌中阳性率分别达到75.0％ (45/60)和68.3％ (41/60).统计学分析显示NEAT1和NEAT2在子宫颈癌组织中的表达与肿瘤的分期相关(P＜0.05),与癌细胞浸润深度明显相关(P＜0.01),与肿瘤的淋巴结转移相关(P＜0.01);与年龄、组织学类型无关.结论:人子宫颈癌细胞异常表达NEAT1和NEAT2,且与肿瘤分期、浸润和转移密切相关,NEAT1和NEAT2可能参与子宫颈癌的发生与发展.%Objective:To explore the expression and its clinical significance of NEAT1 and NEAT2 in different cervical lesion tissues.Methods:The expression of NEAT1 and NEAT2 were measured in 10 cases of chronic cervicitis, 10 cases of high-grade cervical intraepithelial neoplasia and 60 cases of cervical cancer by Real time-PCR assay.Results:The relative of NEAT1 and NEAT2 expression respectively were 0.34 ± 0.10 and 0.29 ± 0.06 in chronic cervicitis,1.18 ±0.32 and 1.37 ± 0.44 in CIN Ⅱ-Ⅲ,6.25 ± 1.58 and 5.96 ± 1.47 in cervical squamous cancer and 5.03 ± 1.12 and 4.96 ± 1.03 in cervical adenocarcinoma tissue.What more, NEAT1 and NEAT2 were high expressions in cervical cancer tissue and the positive rates were 75.0％ (45/60) and 68.3 ％ (41/60) respectively.Statistical analysis showed that the expression of NEAT1 and NEAT2 in cervical cancer tissue were associated with the staging of the tumor(P ＜ 0.05), and significantly correlated with depth of invasion of cancer cells (P ＜ 0.01)and lymph node tumors metastasis (P ＜ 0.01).However, the expression of NEAT1 and NEAT2 were not associated with age of patients and histological type of cervical cancer tissue.Conclusion:NEAT1 and NEAT2 were abnormal expression in human cervical cancer cells and closely associated with staging of tumor, tumor invasion and metastasis.NEAT1 and NEAT2 may be involved in the occurrence and development of cervical cancer.
