WWOX蛋白及PTEN蛋白在支气管肺癌中的表达及临床意义

         

摘要

目的:探讨原发性支气管肺癌组织中WWOX蛋白和PTEN蛋白的表达及与临床病理特征的关系.方法:采用免疫组化SP法检测WWOX蛋白和PTEN蛋白在支气管肺癌组织和癌旁5 cm以上正常肺组织中蛋白的表达情况.结果:WWOX蛋白和PTEN蛋白在肺癌组织中均存在低表达或表达缺失(P<0.05).在男性肺癌患者中WWOX蛋白和PTEN蛋白的阳性表达率均比女性肺癌患者中的明显降低(10.00% vs56.25%,17.50% vs 71.88%,均为P<0.05);有淋巴结转移的肺癌组织中WWOX蛋白和PTEN蛋白的阳性表达率均比无淋巴结转移肺癌组织中低(15.38% vs 48.48%,23.08% vs 63.64%,均为P<0.05).吸烟指数≥400年支的肺癌患者WWOX蛋白和PTEN蛋白阳性表达率均低于吸烟指数<400年支的患者,差异有统计学意义(13.33% vs 42.86%,23.33% vs 54.76%,均为P<0.05).WWOX蛋白表达与肺癌组织分化程度、病理类型、TNM分期不相关(P>0.05).PTEN蛋白在鳞癌组织中的阳性表达率明显高于腺癌组织(60.87%vs 22.22%,P<0.05).PTEN蛋白阳性表达率在肺癌组织Ⅰ-Ⅱ期中的阳性表达率明显高于Ⅲ-Ⅳ期(68.57% vs 16.22%,P<0.05).WWOX蛋白和PTEN蛋白的表达呈正相关.结论:WWOX蛋白和PTEN蛋白在支气管肺癌组织中的表达缺失或低表达,有望为肿瘤诊断及基因治疗提供新的靶点.WWOX蛋白和PTEN蛋白的异常表达在支气管肺癌癌变中起到了协调作用.%Objective:To investigate the expression of WWOX protein and PTEN protein in the tissues of primary lung cancer and their relationship with clinicopathological features.Methods:The protein expression of WWOX and PTEN gene in paraffin specimens of lung cancer and specimens of normal lung tissues were detected by SP immunohistochemistry.Results:WWOX and PTEN protein expression showed low level or absence in lung cancer specimens.The positive rates of WWOX protein and PTEN protein in male lung cancer specimens were significantly lower than that in female lung tissues(10.00% vs 56.25%,17.50% vs 71.88%,all P < 0.05).The positive rates of WWOX and PTEN protein expression in specimens with lymphnode metastasis were both lower than those in tissues without lymphnode metastasis (15.38% vs 48.48%,23.08% vs 63.64%,all P < 0.05).The positive rates of expression of WWOX and PTEN protein were lower in patients whose smoking index ≥400 than those in patients whose smoking index <400,the differences were significant(13.33% to 42.86%,23.33% to 54.76%,P <0.05).WWOX protein expression in lung cancer had no obvious relationship with the degree of differentiation,histological type and TNM stage of lung cancer.PTEN protein expression in squamous cell carcinomas lung tissues was higher than that in adencarcinomas lung tissues (60.87% vs 22.22%,P < 0.05).The positive rate of PTEN protein expression in stage Ⅰ-Ⅱ of lung cancer tissues was significantly higher than that in stage Ⅲ-Ⅳ (68.57% vs 16.22%,P < 0.05).WWOX protein expression was positively related to PTEN protein expression in primary lung cancer.Conclusion:The aberrant expression of WWOX and PTEN may provide a new target for tumor diagnosis and gene therapy.They play synergetic roles in process of carcinogenesis of lung cancer.
