MicroRNA-34 a对多种肿瘤细胞迁移的影响及其作用机制探讨

         

摘要

MicroRNA-34a,a core member of the p53 network,is down-regulated in multiple types of tumors and thus is regarded as a suppressor microRNA which is implicated in tumor stem cells. MicroRNA-34a directly inhibits the expression of various information regulators,cyclins and CDK and etc. ,and further brings about cell proliferation inhibition,cell senescence and apoptosis,migration and invasion inhibition to get the tumor suppression effects. Cell migration is the prerequisite of the tumor cell invasion and metastasis and the significant process of tumor occurring and development. The low expression of microRNA-34a has been found in the tumors of breast,prostate,bladder, stomach,colon,bone and etc. ,which is related to cell migration improving and finally aggravates tumor metastasis and spreading. To study the relation of microRNA-34a and cell migration and the mechanism between them will provide great help in the future tumor therapy. This article reviewed the influence of microRNA-34a on cell migration and the mechanism of microRNA-34a in different tumor cells in recent studies.%MicroRNA-34a是p53信号通路的核心成分,在多种肿瘤中表达下调,被认为是一种抑癌的microRNA,并且与肿瘤干细胞有着密切的联系。MicroRNA-34a可直接抑制多种信息调控因子、细胞周期蛋白的表达,进而抑制细胞增殖、抑制细胞迁移和侵袭、促进细胞衰老和凋亡,从而达到抑癌效果。细胞迁移是肿瘤细胞侵袭和转移的先决条件,是肿瘤发生发展的重要过程。MicroRNA-34a的低表达常见于乳腺癌、前列腺癌、膀胱癌、胃癌、结肠癌、骨癌等,这种低表达往往与细胞迁移能力增强有关,最终加剧肿瘤转移扩散。本文总结了近期关于microRNA-34a对不同种类肿瘤细胞迁移的影响及其作用机制,并进行了分析与归纳。
