不同浓度胰岛素样生长因子-1对人脐带间充质干细胞诱导成脂后生长状态的影响

         

摘要

Objective To observe the change of optical density of human umbilical cord mes-enchymal stem cells (hUCMSCs) during differentiating into the adipocytes in the condition of insulin-like growth factor-1 (IGF-1) with different concentrations and to elucidate the suitable concentration of IGF-1 in adipocyte induction. Methods hUCMSCs were separated and cultivated in vitro and hUCMSCs were induced to adipocytes. The third passage cells were randomly divided into five groups. The induced culture medium was interfered with different IGF-1 concentration, respectively 40, 60, 80, 100, 120 ng/mL. Then, the 3-(4, 5)-dimethylthiahiazo(-z-yl)-3,5-diphenytetrazoliumromide testing was conducted on the five sample groups to measure the optical density (D). On the 9th day, the samples were stained by Oil red O. Results There was no significant difference among the cells growth in five groups. The difference in D showed no statistical meaning. The lipids were all formed in the five sample groups. The number of lipids in80, 100 and 120 ng/mL groups were obviously more than other two groups (P <0.01). There was no difference among the 80, 100 and 120 ng/mL group. Conclusion Different concentration of IGF-1 has no significant effect on human umbilical cord mesenchymal stem cells. But IGF-1 with high concentration can effectively induce adipogenic differentiation of hUCMSCs and 80 ng/mL is the most proper concentration.%目的 探讨不同浓度胰岛素样生长因子-1 (IGF-1)对人脐带间充质干细胞(hUCMSCs)诱导成脂后生长状态的影响.方法 培养hUCMSCs并传代,将第3代hUCMSCs随机分为5组,成脂肪诱导24h后,分别加入40、60、80、100、120 ng/mL的IGF-1,对5组样本进行四甲基偶氮唑盐检测,测量光密度.于第9天行油红O染色.结果 各组细胞生长趋势一致,光密度值之间的差异无统计学意义.5组细胞均有红色脂滴形成,80、100、120 ng/mL组成脂数量明显多于40和60 ng/mL组(P <0.01); 80、100、120 ng/mL组3组之间成脂数量上的差异无统计学意义.结论 不同浓度IGF-1对hUCMSCs诱导成脂后的生长状态无明显影响,但较高浓度的IGF-1能明显促进hUCMSCs向脂肪细胞分化,且以80 ng/mL为最适宜浓度.
