高原地区成年藏绵羊和小尾寒羊睾丸组织eNOS的差异表达

         

摘要

[目的]研究内皮性一氧化氮合酶(endothelial NOS,eNOS)在高原地区成年藏绵羊和小尾寒羊睾丸组织中的定位及分布特征.[方法]采用免疫组化 SP 法结合半定量法描述染色结果分布密度,蛋白免疫印迹(Western blot,WB)分析比较.[结果]eNOS免疫组织化学分布密度表明,小尾寒羊睾丸精子细胞和精原细胞强阳性表达,而在藏绵羊精子和精原细胞内无明显表达;小尾寒羊间质细胞(leydig cell)和支持细胞(sertoli cell)为中等阳性表达,在藏绵羊弱阳性表达.eNOS在藏绵羊及小尾寒羊小血管均为阳性表达.Western免疫印迹显示 eNOS抗体有较好的反应性,且小尾寒羊睾丸组织蛋白反应性强于藏绵羊睾丸组织提取蛋白.[结论]高原环境中,eNOS可参与调节高原地区绵羊睾丸局部血管舒缩以适应环境温度变化;小尾寒羊生精上皮 eNOS表达较强可能是高寒环境中精子生成过程中的低氧应激反应的一个方面.%[Objective]To study the location of eNOS in the testicular tissues of adult plateau Tibetan sheep and Small-Tail Han sheep from plateau area.[Method]The immunohistochemical SP method,West-ern blot combined with semi quantitative analysis were used to describe the distribution density.[Result]The Immunostaining analysis showed that eNOS were expressed strongly in the sperm cell and spermato-gonial in testicular tissues of Small-Tail Han sheep but no obviously expression in the adult plateau Tibet-an sheep,and also the eNOS expressed midium strongly in the Leydig cell and Sertoli cell of the Small-Tail Han sheep and weakly presented in both cells in adult plateau Tibetan sheep testis.In addition,the positive expression in small blood vessels was strong in both kind of sheep.The Western blot showed that reactions of eNOS antibody were obviously,and the protein reactions from testis tissue of Small-Tail Han sheep were stronger than that of Tibetan sheep testis.[Conclusion]The study suggests that eNOS would participate in vasomotor regulation for oxygen intake of microcirculation in the high altitude hypoxia environment.The strong expression of eNOS in the Small-Tail Han sheep's testicles may be one of the aspects of testicular hypoxia stress reaction and remains to be further research.
