头孢呋辛对哮喘儿童外周血单个核细胞Th1/Th2平衡的影响

         

摘要

目的 研究头孢呋辛对哮喘儿童外周血单个核细胞Th1/Th2平衡的影响.方法 采用流式细胞术检测哮喘和健康儿童外周血单个核细胞IFN-γ和IL-4水平,以及哮喘儿童外周血单个核细胞经头孢呋辛体外干预后的IFN-γ和IL-4水平.结果 与健康儿童相比,哮喘患儿外周血单个核细胞的IFN-γ和IFN-γ/IL-4比值降低,差异有统计学意义(P<0.05);哮喘患儿外周血单个核细胞在体外与头孢呋辛( 100 mg/L)孵育48 h后,IL-4水平升高,差异有统计学意义(P<0.05),而IFN-γ的变化无统计学意义(P> 0.05);IFN-γ/IL-4比值则降低,差异有统计学意义(P< 0.01).结论 哮喘患儿外周血单个核细胞以Th2(IL-4)占优势,Th 1/Th2比值平衡失凋;而头孢呋辛更加剧这一倾斜,不利于哮喘治疗.%Objective To investigate the effect of cefuroxime on balance of Thl/Th2 in peripheral blood mononuclear cell in pediatric asthma. Methods The levels of IFN-gamma (Thl-type cytokines) and IL-4 (Th2-type cytokines) in peripheral blood mononuclear cells (PBMC) isolated from whole blood were determined before and after intervention by cefuroxime in vitro by flow cytometry. Results IFN-gamma and the ratio of the IFN-gamma/IL-4 lymphocytes of the asthma group were significantly lower than that in the control group (P < 0.05). After incubation 48 h with cefuroxime ( 100 mg/L) in vitro, the PBMC' s Th2-type eytokine (IL-4) levels were increased significantly (P < 0.05) , and Thl/Th2 in PBMC from asthmatic children was significantly lower (P < 0.01) , whereas IFN--y was not observed with significant changes (P > 0.05). Conclusions Cefuroxime promotes the production of Th2 in PBMC in pediatric asthma, and results in imbalance of Thl/Th2. Children with asthma cannot benefit from the use of cefuroxime.
