目的 检测黏着斑激酶(FAK)、整合素Src、c-Jun氨基末端激酶(JNK)在结直肠癌组织中的表达,并探讨其与结直肠癌临床病理指标的相关性.方法 采用免疫组织化学SP法检测68例结直肠癌组织与癌旁组织中FAK、Src、JNK的表达,结合结直肠癌临床病理指标分析相关性.结果 68例结直肠癌标本中,FAK、Src、JNK的表达水平均与癌旁组织差异显著(P值分别为0.012,0.002,0.012).3者的表达水平与肿瘤分化程度呈负相关(P值分别为＜0.001,0.002,0.004;r值分别为-0.346,-0.356,-0.340),FAK表达与淋巴结转移及UICC分期相关(P值分别为0.006和0.013),但与患者性别、年龄、肿瘤的大小及位置无显著相关.FAK蛋白的表达与Src及JNK蛋白的表达呈正相关(r=0.357,P=0.001；r=0.318,P=0.047).结论 FAK、Src、JNK在结直肠癌组织中均呈高表达,与肿瘤分化程度呈负相关,且Src、JNK的表达与FAK呈正相关,提示FAK/Src/JNK信号通路在结直肠癌的发生、发展中起重要作用.%Objective To investigate the expressions of focal adhesion kinase (FAK), Src, c-Jun N-terminal kinase (JNK) in human colorectal cancer,and to study their correlation. Methods SP immunohistoehemical technique was used to detect the expression of FAK,Src and JNK in 68 human colorectal cancer and paracancerous tissue samples. Results The expression of FAK,Src and JNK in colorectal cancer were significantly higher than the adjacent normal liver tissues( P = 0.012, P = 0.002, P = 0.012), and respectively negative correlated with the differentiation degree of cancer cells (P < 0.001, r =-0.346, P =0.002, r =-0.356, P = 0.004, r =-0.340). The positive expression of FAK was correlated with lymph node metastasis and UICC stage (P = 0.006,P= 0.013),but not with gender,age,tumor size,or tumor location. Furthermore,the positive expression of FAK was correlated with the expression of Src and JNK (r =0.357,P =0.001 , r =0.318,P = 0.047). Conclusion FAK,Src and JNK is overexpressed in colorectal cancer samples,which is negatively correlated with the degree of differentiation of cancer cells. Moreover, expressions of Src and JNK are related with FAK expression, which imply that signaling pathway of FAK/Src/JNK plays an important role in carcinogenesis and progression of colorectal cancer.
