参叶缓痴胶囊对模型大鼠慢性脑缺血性认知功能障碍改善作用机制研究

         

摘要

目的:观察参叶缓痴胶囊对慢性脑缺血认知功能障碍模型动物空间辨别力的影响及对脑源性神经营养因子(BDNF)、超氧化物歧化酶(SOD)、丙二醛(MDA)的变化,探讨参叶缓痴胶囊改善认知功能障碍的作用机制。方法采用双血管阻塞(2VO)改良法复制大鼠血管性认知功能障碍动物模型。利用 Morris 水迷宫法对模型动物的学习记忆行为进行测试,以动物逃避潜伏期和空间探索行为为观测指标。酶联免疫法观察脑组织 BDNF、SOD、MDA 等变化情况。结果参叶缓痴胶囊可以缩短模型动物逃避潜伏时间并提高其空间探索能力;提高脑组织中 BDNF 含量,提高脑组织中 SOD 的活性,降低 MDA 的含量。结论参叶缓痴胶囊可以改善慢性脑缺血认知功能障碍模型的认知能力,其机制可能与参叶缓痴胶囊调节脑内神经因子及抗氧化作用有关。%Objective Study the spatial resolution effect of Shenye Huanchi Capsule on model animal with chron-ic cerebral ischemia cognitive dysfunction. Study the function of Shenye Huanchi Capsule in delaying dementia on BD- NF,SOD and MDA. Explore the mechanism of Shenye Huanchi Capsule in delaying dementia on improving cognitive dysfunction. Methods The vessel occlusion(2VO)was modified to establish vasal cognitive dysfunction model in rats. Morris water maze was used to test the learning and memory behavior of rats by measuring the escape latency and spatial probe behavior. The changes of BDNF,SOD and MDA were observed in brains with enzyme-linked immunosor-bent assay(ELISA). Results Shenye Huanchi Capsule in delaying dementia shortened the time of escaping latency of rats and improved the spatial probe ability of rats. Shenye Huanchi Capsule in delaying dementia improved the BDNF concentration and SOD activity and decreased the MDA concentration. Conclusion Shenye Huanchi Capsule in dela-ying dementia improved cognitive ability of rats of chronic cerebral ischemia cognitive dysfunction. The mechanism of Shenye Huanchi Capsule in delaying dementia in improving the cognitive of rats may be correlative with its function on regulating nerve factor and antioxygenation.
