RP-HPLC determination of different parts of the arabica coffee (coffee seed kernel, peel,stems and leaves) in the caffeine content. Column: Agilent Eclips XDB-C18 (Sun, 4.6 mm ×150 mm);mobile phase:methanol-water (30∶70,V/V);flowrate:1mL/min;detectionwavelength:280nm;columntemperature was30℃;Caffeine standard curve equation:Y=11.566 2X+2.982 7, R2=1. Caffeine in 16μg/mL-80μg/mL range with good linearity. Determination of the local introduction of the Arabica Coffee Panzhihua caffeine content of coffee beans 15.56 g/kg, peel content 13.62 g/kg, stems and leaves of the content of 21.19 g/kg. Precision results for the coffee seeds, peel,stems and leaves content of the RSD were 1.01%, 1.11%, 1.59%;Repeatability results for the coffee seeds,peel,stemsandleaves,respectively,incaffeinecontentofRSD0.69%, 1.79%, 1.38%;were lessthan 2%, achieve assay requirements. Coffee seeds, peel,stemsand leavesof caffeine recoverieswere 95.6%-108.4%,94%-103.4%,96.5%-102.6%.% 利用RP-HPLC测定小粒种咖啡不同部位(咖啡豆种仁、果皮和茎叶)中咖啡因含量.色谱柱:Agilent Eclips XDB-C18(Sun,4.6 mm×150 mm);流动相:甲醇-水(30∶70,体积比);流速:1 mL/min;检测波长:280 nm;柱温为30℃;咖啡因标准曲线方程为:Y=11.5662X+2.9827,R2=1.咖啡因在16μg/mL~80μg/mL范围内具有良好的线性关系.测定出攀枝花本地引种的小粒种咖啡豆中咖啡因含量为15.56 g/kg,果皮中含量为13.62 g/kg,茎叶中的含量为21.19 g/kg.精密度实验结果为咖啡豆种仁、果皮和茎叶中含量的RSD分别为1.01%、1.11%、1.59%;重复性实验结果为咖啡豆种仁、果皮和茎叶中咖啡因含量的RSD分别为0.69%、1.79%、1.38%;均小于2%,达到含量测定要求.咖啡豆种仁、果皮和茎叶中咖啡因回收率分别在95.6%~108.4%,94%~103.4%,96.5%~102.6%.
