明胶肽抗中枢疲劳作用机制的研究

         

摘要

This study was designed to observe the changes of DA and 5-HT in hypothalamus,diencephalon, corpus striatum of rats. We want to screen the effect of anti-fatigue active peptides,and the adjustability to content of DA and 5-HT. To develop the high value-added products of pigskin gelatin bioactive peptide ,expanse the scope of application of the product,at the same time to provide experimental data for the method of nutrition recovery. We established animal fatigue model for load swimming training,feed gelatin peptide to mice for nutrition intervention,use the microdialysis technique to extract central neurotransmitters from fatigue animal midbrain,use reversed phase high performance liquid chromatography to detect the changes of monoamine neurotransmitter concentration and calculate the ratio of DA/5-HT. Finally,we evaluated self-made gelatin peptide to delay and moderate of central fatigue. Result of experiment:Compare to control group of feeding double distilled water,DA/5-HT of animal experimental group was 0.84,control group was 0.14. Comparison of these two groups,the experimental group was 600%higher than the control group. Gelatin peptide had obviously increased the content of DA,decreased generation of 5-HT,enhanced the effect of central excitatory,alleviated the central fatigue and enhanced the ability of anti-fatigue sport.%检测饲养明胶肽与蒸馏水的大鼠进行力竭性游泳运动后即刻纹状体、间脑及下丘脑等不同部位的多巴胺和5-羟色胺含量,探讨明胶肽对多巴胺与5-羟色胺含量变化的调节及抗中枢疲劳功效,为开发高附加值的明胶活性肽产品,扩展猪皮产品的应用范围提供理论参考,同时为抗中枢疲劳提供新的营养手段。通过负重游泳训练建立动物疲劳模型,用自制的明胶肽灌胃饲养实施营养干预,用高效液相色谱方法检测不同脑区单胺类神经递质的浓度变化并计算DA/5-HT比值,分析、评价自制明胶肽对中枢递质的调节作用。和灌胃双重蒸馏水的对照组比较,饲养明胶肽的动物实验组比值为0.84,对照组为0.14,两组比较,实验组比对照组高600%,明胶肽具有明显的提高DA含量,降低5-HT生成,增强中枢兴奋性的功效,可以有效缓解中枢疲劳,增强抗中枢疲劳的能力。
