三种低聚果糖检测方法的比较

         

摘要

A High Performance Liquid Chromatography-Evaporative Light Scattering Detection method was compared with Paper Chromatography method and High Performance Liquid Chromatography-Refractive Index Detection method in this study. The comparison result showed that the High Performance Liquid Chromatography-Evaporative Light Scattering Detection method was significantly better than the other two methods. The experimental conditions were showed as follows: Compounds were successfully separated on a Waters NH2 column (4.6 mm) by a gradient elution. Acetonitrile and water were selected as the mobile phase A and B with a flow rate of 1.0 mL/min. The parameters of ELSD were set as follow:drift tube temperature was 85℃, air carrier gas flow rate was 2.0 L/min. The result showed that good separation effect was obtained and the recoveries were ranging from 97.73%to 99.39%. The intra-day precision (RSD) was less than 1.2%and the within-day precision (RSD) was less than 2.8 %. The method showed good separation effect, accuracy, reproducibility, and suitable for the detection of fructooligosaccharides in practice.%通过对纸层析法,高效液相色谱-示差检测法和高效液相色谱-蒸发光散射检测法三种低聚果糖检测方法进行对比研究.结果发现,高效液相色谱-蒸发光散射检测法分离度高,重现性和准确性好,操作相对简单易行,优于其它两种方法.该方法采用氨基柱,流动相为乙腈与水,采用梯度洗脱,流速设定1.0 mL/min.ELSD的漂移管温度设定85℃;载气为空气,流速2.0 L/min.低聚果糖各组分的回收率范围为97.73%~99.39%,日内精密度<1.2%;日间精密度<2.8%.高效液相色谱-蒸发光散射检测法简便快捷,准确性,重现性好,更适合于低聚果糖的检测.
