高效液相色谱同时测定葡萄酒中合成色素

         

摘要

A high performance liquid chromatographic method was developed for the simultaneous determina-tion of three synthetic colors(tartrazine,amaranth and ponceau) in wine.Wine samples were pretreated by solid phase extraction. The analysis column was Eclipse XDB-C18 (5μm,4.6 mm ×150 mm i. d),the mobile phase was 0.03 mol/L ammonium acetate solution and methano1with gradient elution,the flow rate was 0.8 mL/min.The column temperature was 45℃,and the injection volume was 10μL, the eluate was quantitated by high perfor-mance liquid chromatography with diode-array detector. Detection wavelengths were set at 230, 425, 515 nm, respectively.The qualitative analysis was based on chromatographic retention time,and the quantitative analysis was performed by the standard curve method of peak area. Under optimized conditions, the linear ranges of three colors were in the range of 0-100 (μg/mL) with correlation coeficients more than 0.999. On the basis of S/N=3,the limits of detection (LODs) were 0.53μg/L for tartrazine, 0.64μg/L for amaranth and 0.80μg/L for ponceau. The limits of quantification(LOQs) were from 1.77 to 2.67μg/L(signal-to-noise ratio 10:1). The average recover-ies ranged from 89.30%to 103.93%with the relative standard deviations (RSD)(n=6) less than 5%.This method was accurate, sensitive and rapid, it has been successfully applied to the determination of illegally added synthetic colors in wine.%建立高效液相色谱法同时测定葡萄酒中柠檬黄、苋菜红和胭脂红3种人工合成色素的方法.样品经固相萃取后,采用Eclipse XDB-C18(5μm,4.6 mm×150 mm i.d)反相色谱柱分离,以0.03 mol/L乙酸铵-甲醇为流动相进行梯度洗脱,柱温45℃,流速0.8 mL/min,进样量10μL,采用二极管阵列检测器进行检测.检测波长分别为230、425、515 nm.以目标物的色谱保留时间进行定性,以色谱峰的峰面积用标准曲线外标法进行定量.在优化的色谱条件下,3种色素的线性范围为0~100(μg/mL),相关系数均大于0.999.以S/N=3确定各检出限,柠檬黄、苋菜红和胭脂红的检出限分别为0.53、0.64、0.80μg/L,定量限为1.77μg/L~2.67μg/L(S/N=10).各目标物的加标回收率为89.30%~103.93%,相对标准偏差均小于5%(n=6).方法准确、快速、灵敏,适用于葡萄酒中非法添加合成色素的检测.
