高效液相色谱-电喷雾串联质谱法鉴定芍药花色苷

         

摘要

目的 建立高效液相色谱-二极管阵列检测器/电喷雾质谱联用技术研究芍药花中的花色苷类化合物的成分和含量的方法.方法 首先用含0.1%盐酸的甲醇溶液提取芍药中的花色苷,经XDA-7大孔树脂纯化后,用高效液相色谱-电喷雾串联质谱(HPLC-DAD-ESI-MS2)进行紫外-可见光谱和质谱分析.结果与结论 鉴定出4种花色苷,经紫外-可见光谱、质谱和文献报道综合分析,确定了芍药花中的主要花色苷是芍药素-3,5-二葡糖苷,含量为77.14%.含量较低的三种花色苷为矢车菊素-3,5-二葡糖苷,芍药素-3,5-乙酸酰二葡糖苷,飞燕草素-3-葡糖苷,含量分别为2.68%,9.78%,9.07%.%Objective The compositions and contents of anthocyanins in the flowers of Paenoia lactiflora were detected by high performance liquid chromatography (HPLC) equipped with a Inertsil ODS-SP C 18 column and a DAD detector and electrospray ionization tandem mass spectrometry (ESI-MS/MS).Methods First the anthocyanins were extracted using methanol containing 0.1% HCl solution,then purificated with XDA-7 macroporous resin and eventually analysised with high performance liquid chromatography-mass spectrometry (HPLC-DAD-ESI-MS2),and four anthocyanins were separated and identified.Results and Conclusion The main anthocyanin in the flowers of Paeonia lactiflora was peonidin-3,5-di-O-glucoside,and the content was 77.14 %.And others were cyanidin-3,5-di-Oglucoside(2.68%),peonidin-3,5-di-O-acetic acid glucoside(9.78 %),delphinidin-3-glucoside(9.07 %).
