我国3株登革2型病毒E基因序列测定及毒力基因位点分析

         

摘要

Aim To determine the E protein genomic sequences and find thegenetic loci which may be responsible for virulence of three dengue 2 virus strains isolated in China.Methods The complete genomic sequences of the E gene of three dengue 2 virus strains were determined by RT-PCR. Results Nucleotide and amino acid homology was more than 95% among the E protein of DEN2-04, 43, 44. Four amino acid substitutions confer red charge or side chain polarity change between DEN2-04 and DEN2-43, 44. Conclusions E-64 and E-203 are the genetic loci which may be responsible for virulence of DEN-2.%目的 测定我国3株临床症状、乳鼠神经毒力不同的登革2型病毒流行株E基因序列并找到可能与毒力相关的基因位点。方法 运用RT-PCR法测定我国3株登革病毒的E基因序列。结果 DEN2-04、43、44株的核苷酸及氨基酸同源性均在95%以上,DEN2-04与DEN2-43、44共有4个氨基酸差异引起极性或电荷的变化。结论 E64、E203很可能是与毒力相关的基因位点。
