头痛宁胶囊对偏头痛大鼠中脑P物质mRNA表达的影响

         

摘要

目的:观察头痛宁胶囊对偏头痛大鼠中脑P物质(Substance P,SP) mRNA表达的影响.方法:健康Wistar大鼠24只,随机分4组:A组:生理盐水对照组,B组:生理盐水+偏头痛组,C组:头痛宁胶囊对照组,D组:头痛宁胶囊+偏头痛组.A、B组灌胃给予生理盐水2 ml/d,C、D组灌胃给予头痛宁胶囊0.375 g/kg/d,4组均连续给药7d.给药结束后,B、D组给予硝酸甘油制备偏头痛模型,A、C组给予同剂量花生油对照,2h后处死大鼠,取中脑.应用实时定量PCR技术,对各组大鼠中脑P物质mRNA进行绝对定量.结果:大鼠中脑P物质mRNA拷贝数A组、C组和D组高于B组(P<0.05),A组、C组和D组之间无显著性差异(P>0.05).结论:头痛宁胶囊通过上调偏头痛大鼠中脑P物质表达,发挥治疗偏头痛的作用.%Objective: To investigate the effects of Toutongning capsule on the expression of substance P (SP) mRNA in the midbrain in migraine rats. Methods: The 24 adult Wistar rats were randomly divided into 4 groups: the NS control group (group A), the NS + migraine headache group (group B), Toutongning capsule control group (group C), Toutongning capsule + migraine headache group (group D). In the group A and B, NS (2 ml/d) was administered intragastrically for 7 days prior to treatment with peanut oil or nitroglycerin. In the group C and D, Toutongning capsule (0.375 g/kg/d) was administered intragastrically for 7 days prior to treatment with peanut oil or nitroglycerin. In group D and B, the migraine headache was induced by subcutaneous injection of nitroglycerin. In group A and C, the rats were injected with the same dose of peanut oil. Two hours after peanut oil or nitroglycerin injection, the rats were anesthetized with 10% chloral hydrate (0.3 ml/100 g). Six rats from each group were decapitated, and the midbrains were removed and detected SP mRNA expression by real-time quantitative PCR. Results: The expression of SP mRNA in group A, C and D was higher than that in group B (P < 0.05). No significant difference in SP mRNA expression among group A, C and D (P > 0.05). Conclusion: Toutongning capsule can efficiently inhibit migraine headache by up-regulating SP mRNA expression in the midbrain.
