Objective To contribute theoretically to the selection of the optimal concentration of the photosensitizer through the determination of the level at which C.albicans generate protoporphyrin IX (PpIX) under different concentrations of the photosensitizer and of the bacteriostatic rate of 5⁃amino ketones pentanoic acid photodynamic therapy (5⁃ALA PDT).Methods C.albicans suspen⁃sion was prepared and incubated with ALA in the dark.The generation of PpIX was observed under a laser confocal microscope.The inhibitory rate of 5⁃ALA PDT on C.albicans was determined by MTT method.Results After dark incubation, C.albicans and ALA produced fluorescent PpIX. ALA concentration was positively correlated with the intensity of PpIX. When drug concentration reached300mmol/ L,the intensity of PpIX strength ceased to increase.There was no fluorescent material in control group.Conclusions The inhibitory effect of ALA PDT on C.albicans was closely related to the concentration of the photosensitizer,which provides data for treatment of C.albicans infection.%目的:通过测定白念珠菌内不同光敏剂浓度下生成原卟啉 IX (PpIX)的水平及5⁃氨基酮戊酸光动力疗法(5⁃ALA⁃PDT)对白念珠菌的抑菌率,为临床选择最佳光敏剂浓度提供理论依据。方法制备白念珠菌悬液,与 ALA 避光孵育,采用激光共聚焦显微镜观察 PpIX 产生情况,MTT 法测定5⁃ALA⁃PDT 对白念珠菌生长的抑制率。结果白念珠菌与 ALA避光孵育后有荧光物质 PpIX 产生,ALA 光敏剂浓度与 PpIX 强度呈正相关,当 ALA 浓度到达300 mg/ mL 时,PpIX 强度将不再增加。对照组无荧光物质出现。结论 ALA⁃PDT 对白念珠菌抑制效应同光敏剂浓度密切相关。这为临床 ALA⁃PDT 治疗白念珠菌疾病提供了理论依据。
