大鼠实验性牙周炎对胰岛的影响

AIM : To investigate the effect of ligaLure-induced periodontitis on pancreatic islets in SD rats.METHODS : Twenty five SD rats were randomly divided into periodontitis group( n = 15 ) and control group( n = 10).Bilaeral maxillary second molars of penodontitis group were ligated to induce periadontitis.Blood samples were collected at baseline and in the week.Rats were sacrificed in week12, maxilla and pancreas were dissected for histological analysis.The deqre of alveolar bone resorption was recorded.Cyclooxygenase( COX)-2 expression in pancreatic islets was determined by immunostaining.RESULTS : Serum tumor necrosis factor ( TNF)-a level of periodontitis group and control group were( 10.02 ± 2.16) pg/mL and ( 7.64 ± 1.59) pg/mL in week 12 respectively , with significant difference between the two groups (P＜0.05).However, serum insulin level, histological analysis of pancreas and COX-2 expression level in pancreatic islands did not present obvious difference between the two groups.CONCLUSION: Destruction of pancreatic islets was not able to he observed in the periodontitis rat model constructed in the present study.%目的:通过观测实验性牙周炎大鼠胰岛组织形态、检测外周血胰岛素水平和胰岛环氧化酶(cyclooxygenase,COX-2)表达情况,探讨慢性牙周炎对胰岛的影响.方法:将25只SD大鼠随机分为对照组10只和慢性牙周炎组15只,慢性牙周炎组行双侧上颌磨牙结扎法建立牙周炎模型.基线和12周采集外周血,12周处死大鼠,取颌骨和胰腺组织标本做切片,观察牙周和胰腺的组织学改变,测量牙槽骨吸收高度,并采用免疫组织化学法检测胰岛COX-2表达情况.结果:12周时牙周炎组外周血中肿瘤坏死因子(tumor necrosis factor-a,TNF-a)达(10.02±2.16)pg/mL,较对照组(7.64±1.59)pg/mL有明显升高(P<0.05).但牙周炎组的血清胰岛素水平、胰岛形态和COX-2的表达水平均未见明显差异.结论:中等程度慢性牙周炎尚不足以造成大鼠胰岛组织形态和功能的改变.
