TGF－β1在人慢性根尖周病巨噬细胞上表达的研究

摘要

目的：观察人不同类型慢性根尖周病组织中巨噬细胞（MPs ）的分布情况及转化生长因子－β1（TGF－β1）在巨噬细胞上的表达情况，探讨巨噬细胞TGF－β1在根尖周疾病中的作用机制。方法：将75例受试者标本按根尖周病分类标准分为：正常对照组、尖周肉芽肿组、尖周囊肿组（n＝25）。标本经100 g／L中性福尔马林溶液固定48 h，脱水、包埋、制备组织切片、HE染色，光镜下观察各组根尖周标本的组织学变化；免疫荧光双染色法（DIF），荧光显微镜及共聚焦显微镜下分别观察TGF－β1－CD14（MPs标记蛋白）双阳性细胞在各组根尖周组织中的表达情况。结果：与正常对照组相比，人慢性根尖周病组织中TGF－β1－CD14双阳性细胞密度（cells／mm2）明显升高（P＜0．05）；且根尖囊肿组 TGF－β1－CD14双阳性细胞密度显著高于根尖肉芽肿组（P＜0．05）。结论：TGF－β1在人不同类型慢性根尖周病组织中巨噬细胞上有不同程度的表达；TGF－β1－CD14双阳性细胞可能在人慢性根尖周病的病变过程，尤其在根尖周囊肿的纤维形成及组织修复过程中发挥重要作用；巨噬细胞M2型可能在根尖周组织修复过程起关键作用。%AIM:To observe the expression of TGF-β1 of macrophages in the periapical tissues with differ-ent types of human chronic periapical diseases.METHODS:75 specimens from the subjects were divided into 3 groups(n=25 ),including healthy control,periapical granuloma and periapical cyst.All specimens were examined by HE staining and double-immunofluorescence for the identification of TGF-β1-CD14 (MPs)double-positive cells in periapical tissues.RESULTS:Compared with the healthy controls,the densitiy (cells /mm2 )of TGF-β1-CD14 double-positive cells was significantly higher in the tissues of chronic periapical diseases (P<0.05 );and that in periapical cyst group was significantly higher than in periapical granuloma (P<0.05).CONCLUSION:TGF-β1 -CD14 double-positive cells may be involved in the pathological process of human chronic periapical diseases,espe-cially in the process of fibrous formation and tissue repair of periapical cysts.
