钛网三维支架表面纳米管形貌的制备及其体内成骨诱导能力的研究

         

摘要

AIM:To explore the effects of porous titanium with nanotubes on bone formation in vivo. METHODS:Porous titanium were fabricated by vacuum diffusion bonding and the nanotubes were formed by anodic oxidation with the voltage of 20 V and 25 V respectively.The surface morphology of the samples was observed by SEM. 1 2 samples with nanotubes on the surface(experimental group)and 1 2 samples without nanotubes(control group)were implanted in the defects of femur condylus of 1 2 rabbits with sequential fluorescent labeling.The samples with sur-rounding bone tissue were harvested after 1 and 3 months respectively.The hard tissue sections were prepared and bone formation was examined by Micro -CT.RESULTS:Nanotubes with the diameters of 80 nm were fabricated by anodic oxidation with the voltage of 25 V on the surface of the porous titanium.Fluorescent labeling results showed that after 1 month,the experimental group exhibited higher osteogenic ability than the control group.Micro -CT examination showed that after 1 month,the bone volume /total volume (BV /TV)and trabecular number (Tb.N)of the experimen-tal group were higher than those of control group (P <0.05),while the trabecular space (Tb.Sp)of the experimental group was smaller (P <0.05 ).No difference was observed between the 2 groups (P >0.05 )after 3 months. CONCLUSION:Porous titanium with nanotubes may improve osteogenetic ability at early stage of bone healing and the integration of titanium with surrounding bone.%目的:观察钛网三维支架表面构建的纳米管形貌对其体内成骨能力的影响。方法:采用钛网扩散连接的方法构建三维支架,并分别在20、25 V 电压下通过阳极氧化法在其表面构建纳米管形貌。SEM观察各组材料的表面形貌和结构后,植入新西兰兔双侧股骨外髁部骨松质内,定期对各动物进行序列荧光标记。分别于植入后1、3个月取材,用 Micro -CT 扫描分析各组的骨形成量;用硬组织切片观察各组支架表面及内部骨形成情况。结果:20 V 电压在钛网支架表面形成纳米凹坑,25 V 电压可制备出管径约为80 nm 的纳米管结构。植入动物体内后荧光标记显示,实验组1个月时即表现出成骨活性。CT 扫描分析显示,植入1个月时实验组的骨体积分数(BV /TV)、骨小梁数目(Tb.N)均明显高于对照组(P <0.05),而骨小梁间隔(Tb.Sp)则明显小于对照组(P <0.05);植入 3个月后两组各项指标均无显著性差异(P >0.05)。结论:钛网三维支架表面纳米管形貌可以增强材料表面的早期成骨能力,并能促进材料与骨组织的结合。
