Nd:YAG激光对牙龈卟啉单胞菌的杀菌作用探讨

         

摘要

目的:观察Nd:YAG激光对牙龈卟啉单胞菌(Porphyromonas gingivalis,P.gingivalis)的杀菌效果.方法:培养P.gingivalis并置备其生物膜.结晶紫染色后观察功率为6 W的Nd:YAG激光不同照射时间对浮游状态下P.gingivalis成膜能力的影响;荧光染色后分别用激光共聚焦显微镜、扫描电镜观察Nd:YAG激光不同照射时间对P.gingivalis单菌种生物膜活力和结构的影响.结果:Nd:YAG激光照射35 s时,对浮游状态下P.gingivalis的成膜能力有抑制作用;照射45 s时,对生物膜状态下P.gingivalis活力产生抑制.生物膜的结构及生物膜中细菌的结构,随着光照时间延长受到破坏的程度不断增加,主要破坏形式为菌体的汽化消失.结论:Nd:YAG激光能够抑制浮游状态下P.gingivalis的成膜能力,对单一菌种生物膜中的细菌活力和细菌结构产生影响,表明Nd:YAG激光可用于牙周炎辅助治疗.%AIM:To investigate the antibacterial effects of Nd:YAG laser against Porphyromonas gingivalis ( P. gingivalis) in vitro. METHODS:P. gingivalis were in vitro cultured and P. gingivalis biofilm was prepared. The cultured P. gingivalis were treated by Nd:YAG laser at 6 W for 0~60 s respectively. The biofilm formation ability of planktonic P. gingivalis was observed by crystafl violet test; the viability of P. gingivalis cells in mature biofilms was detected by confocal Laser scanning microscopy ( CLSM);the morphology of mature biofilms was observed by scanning electron microscopy ( SEM) . RESULTS:The biofilm formation ability of P. gingivalis was limted under 35 s laser radi-ation. The viability of P. gingivalis in mature biofilm went down under 45 s iradiation. The deformation of mature bio-film and the deconstruction of becterium became distincted with the iradiation time increased. The major destructional form was evaporation. CONCLUSION:Nd:YAG laser may inhibit of biofilm formation of planktonic P. gingivalis and destroy the construction of the mature biofilm and bacteria.
