FOX03a基因转染对血管损伤后新生内膜增生的影响

         

摘要

Objective To study the inhibitional effects of transcription factor foxO3a gene transfection on neointimal hyperplasia after balloon injury in rats. Methods The common carotid catheter injury model in rats was established. A tote of 54 male wistar rats were divided into three groups. Immediately after angioplasty, pECE-HA-FOXO3a-TM vector and pECE ( -)vector respectively mediated by Lipofectamine 2000 or PBS was transfected into the injured segmet of the common carotid artery for local therapy. Three days late,the transfected vasculars were obtained to detect expression of toltal FOXO3a and HA in cultured smooth muscle cells. And then intimal-media ratio and neointimal hyperplasia were measured at different time. Results The transfected common carotid arteries were confinned the expression of HA in cultured smooth muscle cells. 1 and 3 months after transfection ,the intimal-media ratio was signigicantly reduced in FOXO3a-TM transfected group than that of PBS group and pECE ( - ) transfected group (P < 0.0l ). Conclusion FOXO3a-TM gene transfection might inhibit strongly neointima hyperplasia after balloon injury and reduce arterial restenosis in rats.%目的 观察FOX03a三倍突变体(FOX03a-TM)基因体内局部转染对大鼠血管损伤后新生内膜增生的抑制作用.方法:实验建立大鼠颈动脉球囊损伤模型,在建立模型基础上,随机分成空白对照组、质粒对照组和FOXO3a-TM组.损伤术后,分别将PBS、脂质体Lipofectamine 2000介导pECE(一)和pECE-HA-FOXO3a-TM体内转染至以上三组大鼠损伤血管壁.转染3d后培养并鉴定损伤局部平滑肌细胞,并以Western blotting法检测各组损伤及转染后血管平滑肌细胞中标记蛋白HA以及FOX03a总蛋白的表达,以证实HA-FOX03a-TM的有效表达,同时观察转染后不同时期新生内膜增生的影响.结果 三组颈总动脉血管平滑肌细胞均有FOX03a表达,其中FOX03a-TM组表达HA,空白对照组、质粒对照组不表达,证实HAFOX03a-TM的有效表达.以上三组大鼠转染后1和3个月,FOX03a-TM组内膜/中膜面积比值比空白对照组、质粒对照组显著降低(P<0.01).结论 FOXO3a-TM基因体内转染损伤后血管可以增加活性转录因子FOX03a表达,抑制血管新生内膜增生,减少再狭窄发生率.
