Objective To investigate the effects of Jiawei Baiteng extracts on the pharmacokinetics of cefaclor in rats,and thereby to discuss the rationality of the combination therapy.Methods Twelve rats were randomly divided into two groups:cefaclor group and cefaclor+Jiawei Baiteng extracts group,with the same dosage of cefaclor (125mg/kg).Blood samples were collected from the retinal venous plexus of each rat in the two groups after a single oral dose.The plasma samples were pretreated with methanol,and were then examined by HPLC-TQ-MS/MS assay.The pharmacokinetic parameters of cefaclor in rats were calculated by DAS software (version 2.0) and analyzed statistically.Results The main pharmacokinetic parameters of cefaclor in cefaclor group were as follows:Cmax=(31.58±9.72)μg/mL,AUC=(37.48±14.34)μg/(mL·h),t1/2=(0.94±0.10)h,Tmax=(0.50±0.16)h,C1=(3.95±2.12)L/(h·kg),while those in cefaclor+Jiawei Baiteng extracts group were Cmax=(27.76±5.62)μg/mL,AUC=(35.53±11.62)μg/(mL·h),t1/2=(1.35±0.20)h,Tmax=(0.83±0.13)h,C1=(3.69± 1.13)L/(h·kg).The parameters of t1/2 and Tmax of cefaclor were significantly extended when co-administered with Jiawei Baiteng extracts(P＜0.05).Conclusion Jiawei Baiteng extracts prolonged the action duration of cefaclor in rats by delaying its elimination,which suggested that the combination therapy might be reasonable.%目的 研究加味败藤浸膏对大鼠体内头孢克洛药动学的影响,探讨联合用药的合理性.方法 大鼠随机分为2组:头孢克洛单用组和头孢克洛+加味败藤浸膏联用组,头孢克洛剂量均为125 mg/kg.各组大鼠给药后于不同时间眼底静脉丛取血,血浆经甲醇预处理后进行HPLC-TQ-MS/MS分析.DAS统计软件计算头孢克洛的药动学参数,并对其药动学参数进行统计学分析.结果 单用组头孢克洛的主要药动学参数分别为:Cmax=(31.58±9.72)μg/mL,AUC=(37.48± 14.34)μg/(mL.h),t1/2=(0.94±0.10)h,Tmax=(0.50±0.16)h,C1=(3.95±2.12)L/(h·kg);联用组头孢克洛的主要药动学参数分别为:Gmax=(27.76±5.62)μg/mL,AUC=(35.53± 11.62)μg/(mL.h),t1/2=(1.35±0.20)h,Tmax=(0.83±0.13)h,C1=(3.69±1.13)L/h/kg.同时服用加味败藤浸膏时,大鼠体内头孢克洛的t1/2与Tmax显著性延长(P＜0.05).结论 加味败藤浸膏可延缓头孢克洛的消除速度,延长其体内持续作用时间,联合用药具有一定合理性.
