Objective: To construct murine model of Candida glabrata ( C.glabrata) vaginitis.Methods: After ICR female mice were intravenously injected with 160 mg/kg streptozotocin (SZ) or intraperitonealy dexamethasone 0.5 mL, the mice were subcutaneously injected with estradiol to induce pseudooestrus.And then, vaginal stationary -phase blastospore of Candida glabrata suspension (5× 108 cfu/mL) was injected.The amount of Candida glabrata in vaginal lavage fluid was analyzed and vaginal pathological changes were observed.Results: Compared with the control group, there was high load of C.glabrata in the culture of lavage fluid on day 7 after inoculation; vaginal tissue from C.glabrata- inoculated mice contained large amount of blastospores in the superficial mucoss and model persisted for 2 weeks.Conlusion: Use of immunosuppressive vaginal estrogenization and high concentration of C.glabrata inoculation can construct a stable model of C.glabrata vaginitis.This model can provide a means to study the pathogenesis of mucosal C.glarata infections.%目的:构建光滑念珠菌性阴道炎的小鼠模型.方法:采用ICR雌性小鼠,预先静脉注射链脲菌素或腹腔注射地塞米松,再皮下注射苯甲酸雌二醇以造成小鼠的假发情,然后小鼠阴道内注射约5×108cfu/mL光滑念珠菌稳态芽生孢子并进行阴道灌洗液的真菌载量分析以及阴道组织病理观察.结果:与对照组小鼠比较,实验组小鼠阴道灌洗液的真菌载量分析显示自第7 d开始可分离出高水平的菌量,并延续到接种后21 d;阴道组织病理结果发现自第7 d至实验结束均可见阴道黏膜浅层有芽生孢子生长,并有炎细胞浸润.结论:小鼠胰岛功能丧失及全身免疫抑制,加以雌激素化及阴道内高浓度菌悬液接种可以构建稳定的光滑念珠菌阴道炎模型,此动物模型可用于念珠菌性阴道炎的相关研究.
