目的::检测2型单纯疱疹病毒( HSV-2)感染后HaCaT细胞分泌IL-17、IL-23 mRNA的水平变化,明确IL-17和IL-23与角质形成细胞( KC)是否参与了HSV-2病毒感染初期的免疫防御机制。方法:用 HSV-2病毒感染HaCaT细胞,采用荧光实时定量PCR检测感染后24 h、48 h、72 h及空白对照组IL-17、IL-23 mRNA的表达。结果: HSV-2病毒感染后72 h HaCaT细胞分泌IL-17 mR-NA的表达量低于对照组,而IL-23 mRNA表达量高于对照组( P<0.05)。结论:在HSV-2感染KC初期,IL-17和IL-23可能参与早期免疫防御反应。%Objective:To detect the changes of IL-17 and IL-23 mRNA levels in HaCaT cells after infec-ted by HSV-2, in order to determine whether or not the IL-17 and IL-23 are involved in the defense immune mechanism to HSV-2 infection. Methods:The levels of IL-17 and IL-23 mRNA in HaCaT cells were detec-ted by Real-time PCR after 24 h, 48 h and 72 h after infected by HSV-2. Results:The level of IL-23 mR-NA was higher than that in the control group after 72 h infected by HSV-2 ( P<0.05) , while the level of IL-17 mRNA was lower than that in the control group (P<0.05). Conclusion: IL-17 and IL-23 may play an important role in the defense immune mechanism of KC to HSV-2 in the early stage of the infection.
