Characterization of local mineral leaching bacterial isolates (Acidithiobacillus ferrooxidans, Leptospirillum ferrooxidans and Acidithiobacilllus thiooxidans) from a gold mine was done using conventional microbiological methods and by sodium dodecyl sulphate polyacrylmide gel electrophoresis (SDS-PAGE). Further characterization of local mineral oxidizing microbes was achieved by 16S rRNA analysis. In this work, we look at differences between local acidophilic bacteria obtained from surface arsenopyrite and underground sulphide gold ores using the transmission electron microscope and restriction enzyme analysis of bacterial 16S rRNA. No apparent differences were observed between bacterial species isolated from either surface or underground ores using amplified ribosomal DNA restriction enzyme analysis (ARDRA) in this study.
